Marburg Virus Glycoprotein Is a Remarkable Virulent Factor Linked to Hemorrhagic Pathology: Evidence from Multimodal Experimental Systems

open access: yesAdvanced Science, EarlyView.
By integrating data from in vitro, ex vivo, and in vivo models, our research identifies the MARV glycoprotein as a remarkable hemorrhagic factor, filling a major gap in this important field. It also provides practical experimental tools for the basic research on viral pathogenesis and applied research aimed at antiviral intervention for hemorrhagic ...

Unveiling Therapeutic Opportunities with Melanoma Patient-derived Organoid Models

open access: yesJournal of Visualized Experiments
With the development of immunotherapy, there is an ongoing need to develop models that can recapitulate the tumor microenvironment of native tumors. While traditional two- and three-dimensional models can offer insights into cancer development and progression, these lack crucial aspects that hinder a faithful mimic of native tumors.

Conformable Microelectrode Arrays Integrated with a Scoop‐Shaped Slide‐Well for Dynamic Electrophysiological Profiling of Patient‐Derived Cardiac Organoids

open access: yesAdvanced Science, EarlyView.
A flexible, ultrathin multi‐channel microelectrode array (MEA) conformally integrated onto a curved slide‐well enables stable electrical interfacing with 3D cardiac organoids. The geometry‐guided self‐alignment allows simultaneous recording of extracellular field potentials and contractile motion from irregular, beating tissues.

BGN/MDK Axis in the Melanoma Tumor Microenvironment Strengthens Tumor Malignancy by Modulating Cancer Cells and Cancer‐Associated Fibroblasts Crosstalk

open access: yesAdvanced Science, EarlyView.
This study reveals that m6A regulators cooperatively upregulate BGN in melanoma, promoting malignancy. Within the tumor microenvironment, CAFs show highest BGN expression. The BGN/MDK axis mediates cancer‐stroma crosstalk, driving normal fibroblast (NF) activation and enhancing the pro‐tumor effect of CAFs, highlighting a promising therapeutic target ...

Gut Mycobiota‐Associated Tryptophan Catabolites Protect Against Metabolic Dysfunction‐Associated Steatotic Liver Disease

open access: yesAdvanced Science, EarlyView.
ABSTRACT Accumulating evidence suggests that the intestinal microbiota participates in the progression of metabolic dysfunction‐associated steatotic liver disease (MASLD) through microbiota‐host interaction. However, the beneficial role of commensal mycobiota in MASLD progression remains poorly understood.

Personalized Human Astrocyte‐Derived Region‐Specific Forebrain Organoids Recapitulate Endogenous Pathological Features of Focal Cortical Dysplasia

open access: yesAdvanced Science
Focal cortical dysplasia (FCD) is a highly heterogeneous neurodevelopmental malformation, the underlying mechanisms of which remain largely elusive. In this study, personalized dorsal and ventral forebrain organoids (DFOs/VFOs) are generated derived from

A 3D In Vitro Model of the Human Hepatobiliary Junction

open access: yesAdvanced Science, EarlyView.
A 3D human organoid platform reconstructs the hepatobiliary junction between primary adult hepatocytes and intrahepatic cholangiocytes. These adult hepatobiliary organoids (aHBOs) support directional bile transport from canaliculi to ductule‐like structures, enable quantitative imaging of junction dynamics, and reveal cell‐type‐specific vulnerabilities

Generation and Maintenance of Patient-Derived Endometrial Cancer Organoids

open access: yesBIO-PROTOCOL
Endometrial cancer (EC) is the leading cause of gynecologic cancer morbidity and mortality in the U.S. Despite advancements in cancer research, EC death rates are increasing, particularly high-grade endometrial cancers. The development of three-dimensional (3D) patient-derived organoid (PDO) models for EC is crucial, as they provide a more accurate ...
