Electrophoresis, 1995AbstractA rapid and sensitive method of detecting pectinesterase activity following electrophoresis or isoelectric focusing in polyacrylamide gels is described. The method uses ruthenium red and requires no addition of substrate when making the gels, thus obviating direct enzyme‐substrate contact during electrophoresis.

Purification and Properties of Apple Pectinesterase
Journal of the Science of Food and Agriculture, 1996Pectinesterase from apple was separated into two fractions by chromatography on Sephadex G-75. The major pectinesterase was purified by cation-exchange chromatography on CM-Sephadex C-50. It had a molecular mass of 36 kDa, and isoelectric point of about 9, similar to one of the major pectin-esterases of lemon.

The experiments were separated into two groups. The first ones were performed at 40°C-90°C, ambient pressure with ultrasonication of 20 kHz. The other ones were carried out at 40°C-90°C without ultrasonication.
