A novel form of pectinesterase in tomato

The pectinesterases in tomato pericarp were fractionated by cation exchange into four forms (A-D). Form A was the most abundant and C the second most abundant. Forms A-C were further purified by gel filtration, and antibodies were raised against A and C.

AbstractMethanol accumulates in axis tissues of maturing soybean seeds, correlating with preharvest seed deterioration. Accumulation of methanol appears to be associated with the enzymic demethylation of pectin methyl esters by pectinesterase (PE; EC 3.1.1.11).

Kinetics of papaya pectinesterase

Abstract Purified papaya (Carica papaya L. var. exotica) pectinesterase (EC 3.1.1.11) was investigated for its activity as a function of NaCl, pH and temperature, and determination of its kinetic parameters. The activity was linear up to 20 min with an enzyme concentration of up to 6.14 μg.

Papaya Pectinesterase inhibition by Sucrose

SUMMARY Pectinesterase (PE) was inhibited by sucrose at the same concentrations that delayed gelation of papaya purée. The inhibitory effect was linear with sucrose concentration throughout the range investigated (up to 50% sucrose).

Action of iodine on the tomato pectinesterase

The organophosphates and carbamates did not inhibit the isolated tomato pectinesterase. Therefore this enzyme cannot be defined as serine-type esterase. The enzyme is inhibited by iodine and the inhibition (irreversible and non-competitive) is dependent on the degree of enzyme purification.

Abstract A rapid assay for inhibition of pectinesterase has been developed to facilitate large scale screening for possible inhibitors. The assay gives quantitative results which can be confirmed by other assay techniques. The assay uses an agar plate in which pectin and an indicator have been incorporated.

Stability studies of papaya pectinesterase

Abstract Stability studies were carried out on the purified papaya ( Carica papaya L. var. exotica ) pectinesterase (EC 3.1.1.11). The enzyme preparation was stable at 4°C in 0·02m sodium phosphate buffer (pH 7·5) solution containing 0·2m NaCl and 0·02% sodium azide and the loss in activity was less than 5% after storage for 1 year.

Pectinesterase extraction from papaya

Pectinesterase (EC.3.1.1.11) was extracted from papaya fruit (Carica papaya L.) var. Exotica. Incubation time, pH and NaCl concentration influenced the extraction process of pectinesterase from papaya fruit. A maximum activity of 6·98 units/min ml was obtained with 2 m NaCl solution, pH 8, and at 5 h incubation time.
