Linkage analysis on complex pedigrees by simulation

Mathematical Medicine and Biology, 1994
A Markovian scheme for simulating the genotypes of individuals in a pedigree subject to constraints imposed by observed phenotypes is used to estimate the logarithm of a likelihood ratio in linkage analysis. The method is illustrated on a fictitious pedigree by comparison with exactly calculated results.

PEDIGREE GENERATION FOR ANALYSIS OF GENETIC LINKAGE AND ASSOCIATION

Biocomputing 2004, 2003
We have developed a software package, SIMLA (simulation of linkage and association), which can be used to generate pedigree data under user-specified conditions. The number and location of disease loci, disease penetrances, marker locations, and marker disequilibrium with a disease locus and with other markers can be controlled.

Pedigree Analysis of Children with Phonology Disorders

Journal of Learning Disabilities, 1992
This study examined 87 pedigrees of individuals with histories of preschool phonology disorders. Results confirmed previous reports that speech and language disorders aggregate in families, with a higher incidence of males affected than females. Significantly more family members with dyslexia and learning disabilities, but not stuttering or hearing ...

Optimal Computation of Probability Functions for Pedigree Analysis

Mathematical Medicine and Biology, 1986
The problem of finding optimal peeling sequences is considered, and a correspondence between sequences and permutations of marriages is established. Several search methods, including random downhill search and the annealing algorithm, are compared. The topology of the space of all peeling sequences is discussed.

Approximate Computation of Probability Functions for Pedigree Analysis

Mathematical Medicine and Biology, 1986
Approximation by factorization of R functions is considered as a way of making the peeling method for calculation of probability functions more efficient and applicable to larger and more complex pedigrees. We show that, in some situations, the method is not approximate but a simplified way of calculating the true probability functions.

Pedigree Analysis of Complex Models

1980
A basic component of any analysis of genetic data on members of a pedigree is the probability that, conditional on the structure of their pedigree and the model for the trait, a set of individuals will have certain phenotypes or genotypes. Risk probabilities required in genetic counseling problems (Binet, Sawyer, and Watson, 1958; Murphy, 1970; Chase ...
