Polyglycerol‐Based Lipids: A Next‐Generation Alternative to PEG in Lipid Nanoparticles for Advanced Drug Delivery Systems

open access: yesMacromolecular Rapid Communications, EarlyView.
Linear polyglycerol is proposed as a PEG‐alternative in mRNA‐loaded lipid nanoparticles to limit activation of anti‐PEG antibodies and reduce immunogenicity. Resulting nanoparticles may provide similar gene delivery capabilities, while preventing binding of anti‐PEG antibodies compared to PEGylated counterparts.

Arsonic Acid Functional Polymers Enable Stabilization of Iron Oxide Nanoparticles in Aqueous Solution

open access: yesMacromolecular Rapid Communications, EarlyView.
Arsonic acid containing diblock copolymers are developed to incorporate arsonic acid moieties for surface anchoring to IONPs, along with water soluble PEG blocks to impart colloidal stability in aqueous environments. The resulting polymer‐coated IONPs are evaluated for stability in physiological media and cytocompatibility.

A Non‐Antigenic Randomized Polyethylene Glycol/Poly(2‐Phenyl‐2‐Oxazine)‐Based Drug Delivery Platform

open access: yesMacromolecular Rapid Communications, EarlyView.
Randomized PEG (rPEG), a conceptually new PEG alternative, is combined with poly(2‐phenyl‐2‐oxazine) (PPheOzi) to establish an rPEG‐b‐PPheOzi‐b‐rPEG micellar drug delivery platform. The PPheOzi block enables high drug loadings, while competitive ELISA reveals a drastic reduction in anti‐PEG antibody affinity of rPEG in comparison to PEG.

PEGylation of lipophilic SN38 prodrug with DSPE-mPEG2000 versus cremophor EL: comparative study for intravenous chemotherapy

open access: yesDrug Delivery, 2019
The lipophilic prodrug of hydrophobic drugs with well-designed molecular structures can form stable pure prodrug nanoparticles (NPs), but rapid NPs aggregation in plasma greatly restricted their direct use for intravenous chemotherapy.

Machine Learning‐Driven Predictive Modeling and Multi‐Objective Exploration of Oxaliplatin‐Loaded Nanocarriers for Enhanced Loading and Encapsulation Efficiency

open access: yesMaterials Genome Engineering Advances, EarlyView.
Integrating ML and multi‐objective optimization enabled efficient, accurate design of nanocarriers with optimized loading and encapsulation efficiencies. ABSTRACT Oxaliplatin (OXA), a critical third‐generation platinum chemotherapeutic, is significantly limited by suboptimal loading capacity and encapsulation efficiency in nanoparticle‐based delivery ...

Hypersensitivity to Excipients in Drugs: An EAACI Position Paper

open access: yesAllergy, EarlyView.
ABSTRACT Drugs contain active pharmaceutical ingredients and excipients, compounds which enhance the pharmacokinetics, stability and palatability of the pharmaceutical formulation. While most drug hypersensitivity reactions (DHR) are caused by active ingredients, excipients may also be involved.

Engineering PEGylated Antibody Fragments for Enhanced Properties and Cancer Detection [PDF]

open access: yes, 2016
3E8 is an antibody that targets Sialyl-Tn, a glycan found exclusively on a subset of cancers called adenocarcinomas. This antibody is particularly interesting to the Magliery Lab because of its use in radioimmunoguided surgery, a modern cancer-detecting ...
