Permeabilization of Cell Membranes
2009In order to detect intracellular antigens, cells must first be permeabilized especially after fixation with cross-linking agents such as formaldehyde and glutaraldehyde. Permeabilization provides access to intracellular or intraorganellar antigens. Two general types of reagents are commonly used: organic solvents, such as methanol and acetone, and ...

Mitochondrial Membrane Permeabilization in Cell Death
Physiological Reviews, 2007Irrespective of the morphological features of end-stage cell death (that may be apoptotic, necrotic, autophagic, or mitotic), mitochondrial membrane permeabilization (MMP) is frequently the decisive event that delimits the frontier between survival and death.

1990
Abstract For many years investigations of cell regulation, and in particular stimulus—response coupling, were limited to studies of intact, viable cells. Details of intracellular mechanisms remained obscure until it was discovered that permeabilized cells, in which the cytosol is directly accessible from the exterior, retain specialized ...

Endocytosis Assays in Intact and Permeabilized Cells
Current Protocols in Cell Biology, 2005AbstractClathrin‐coated pits and vesicles represent the major ports of entry into most eukaryotic cells. As well as performing housekeeping functions (e.g., allowing cells to take up essential nutrients), the endocytic pathway participates in a number of tissue‐specific events such as synaptic‐vesicle recycling, control of morphogen gradients during ...

Digitonin‐Permeabilized Cells Are Exocytosis Competent
Journal of Neurochemistry, 1987Abstract:Release of norepinephrine from PC12 cells can be stimulated by free Ca2+ in micromolar concentrations after permeabilization with 10 μg/ml of digitonin. This release is time and temperature dependent, half‐maximal at 0.3 μM Ca2+, and, after washing out of endogenous ATP, half‐ maximal at about 0.5 mA/ MgATP when exogenously added.

Nuclear Import in Digitonin‐Permeabilized Cells
Current Protocols in Cell Biology, 2000AbstractImport of proteins into the nucleus of a cell is a complex process that can be reconstituted in vitro. Digitonin‐permeabilized cells are washed free of cytosolic factors to provide competent nuclei. Cytosolic factors for import are provided by an extract of Xenopus ovarian cells.

Publisher Summary Permeabilized cells allow measurement of authentic replicative DNA synthesis. They are notable for ease of preparation and a minimal disturbance of the replicative apparatus. The limitation is that there is no initiation of new cycles of DNA replication.

A method for analyzing transcription using permeabilized cells
Analytical Biochemistry, 1984Brief exposure of Friend cells to a buffered hypotonic solution containing 1% Tween 80 caused permeabilization and allowed incorporation of [3H]UTP into RNA. The incorporation was inhibited 85-97% by 20 micrograms/ml actinomycin D and the reaction product was completely hydrolyzed by 0.1 M KOH.

PERMEABILIZED MICROBIAL CELL CATALYSTS
A permeabilized whole cell catalyst and method for conversion (reducing the amount) of a target substrate to one of more product are provided, wherein the catalyst is obtained by incubating the cells with a permeabilization agent selected from a fatty ...

Enzyme assays using permeabilized cells of Neurospora
Analytical Biochemistry, 1979Abstract A procedure is described for the preparation of homogeneous, steady-state cultures of germinated conidia of Neurospora crassa . Permeabilization of such cells has been accomplished by a combination of toluene-ethanol and freeze-thaw treatments. These permeabilized cells have been used for the determination of enzyme activities.
