Macrophage RSAD2 Couples mtDNA Synthesis With a Self‐Amplifying Inflammatory Circuit to Orchestrate Tissue Repair

open access: yesAdvanced Science, EarlyView.
Macrophage RSAD2 dually regulates CMPK2—the rate‐limiting enzyme for mitochondrial DNA synthesis—by suppressing its ubiquitination and promoting its phosphorylation via Csnk2a2 recruitment. This amplifies mtDNA production, which simultaneously activates a cGAS‐STING‐IRF3‐RSAD2 feedforward loop and the NLRP3 inflammasome, driving a self‐sustaining ...

Hyper‐Intense Tumor Peripheral Accumulation of Antibody‐Conjugated Iron Oxide Nanoparticles can Enable Breast Cancer Detection by Magnetic Particle Imaging

open access: yesAdvanced Science, EarlyView.
Tumor‐associated inflammation retains antibody‐targeted or non‐specific antibody conjugated nanoparticles within the tumor microenvironment. Magnetic Particle Imaging (MPI) enables non‐invasive detection and differentiation of tumors from other organs, owing to higher nanoparticle retention by inflammatory cells in the tumor periphery.

Predicting Single‐Cell Perturbation Responses Across Biological Contexts With a Deep Generative Model Integrating Optimal Transport

open access: yesAdvanced Science, EarlyView.
Single‐cell perturbation responses are predicted across held‐out biological contexts using scPILOT, a query‐conditioned two‐stage latent response‐transfer framework. A shared latent representation supports cell‐level response estimation by latent optimal transport, followed by Leiden‐localized query‐specific transfer and adaptive weighting.

Chemotherapy‐Activated GSK3β‐DNMT1 Signaling Upregulates CD47 to Evade Macrophage Phagocytosis and Drive Temozolomide Resistance in Glioblastoma

open access: yesAdvanced Science, EarlyView.
Temozolomide treatment activates GSK3β, driving DNMT1 phosphorylation, destabilization, and CD47 promoter hypomethylation in glioblastoma. This epigenetic shift upregulates CD47, enabling TMZ‐treated GBM cells to evade macrophage phagocytosis, survive chemotherapy, and acquire resistance.

Reversing Macrophage Immunometabolic Dysfunction in Prolonged Infections Via a Glutaminolysis‐Fueled Restoration‐Based Hierarchically Selective Therapeutic System

open access: yesAdvanced Science, EarlyView.
A hierarchically selective immunotherapeutic nanoplatform, mGls@HEV‐MTP, selectively restores macrophage immunocompetence through GLS1‐mediated restoration of glutaminolysis‐fueled anaplerosis. Concurrently, it renders Staphylococcus aureus (S. aureus) more readily recognizable to the metabolically reprogrammed macrophages, thereby facilitating ...
