1. クルマエビ血液中のフェノールオキシダーゼ (PO) 活性の分布を調べるために, 血漿成分と血球成分のPO活性を測定した。その結果, 血漿ならびに血球成分ともにPO活性が認められたが, とりわけ血球中の活性が高かった。2. クルマエビ血球成分中のPO活性の酵素化学的性状について検討した結果, PO活性の反応至適温度は40℃であり, 熱処理または0.5M以上の条件下において著しい活性の低下がみられた。また, 活性はEDTAにより阻害されたが, EGTAによる阻害性は低く, Caイオン依存性ではなかった。3. PO活性はPMSFやDEDTによって阻害されたことから, 活性の反応系にはセリンプロテアーゼや銅酵素の関与が示唆された。

Purification and properties of phenoloxidase from spinach leaves.

A phenoloxidase from spinach leaves was extensively purified to homogeneity. The molecular weight of the purified enzyme was 150,000 by gel filtration, and 40,000 by SDS-polyacrylamide gel electrophoresis. The enzyme contained 2 atoms of copper per 150,000 of molecular weight. The optimum pH of the enzyme action is near 6.5.
