To determine if phospholipase D is present in intact adult islets, we took advantage of the fact that, in the presence of ethanol, this enzyme generates phosphatidylethanol via transphosphatidylation. Extracts of cells prelabeled with [14C]arachidonate, [14C]myristate, or [14C]stearate were analyzed via three TLC systems; the identify of ...

Formation of phosphatidylethanol from endogenous phosphatidylcholines in animal tissues from pig, calf, and goat [PDF]
In the presence of alcohol, phosphatidylcholine (PC) is transformed to the direct alcohol biomarker phosphatidylethanol (PEth). This reaction is catalyzed by the enzyme phospholipase D (PLD) and dependent on substrate availability. As recent methods have

Analysis of the alcohol biomarker phosphatidylethanol by NACE with on‐line ESI‐MS
ELECTROPHORESIS, 2010AbstractPhosphatidylethanol (Peth), a group of aberrant phospholipids formed in cell membranes in the presence of ethanol, has been recently proposed as biomarker of chronic alcohol abuse. The aim of this study was to develop a new analytical method, based on NACE online coupled with a mass spectrometer for the analysis of Peth in blood.

Phosphatidylethanol formation in rat organs after ethanol treatment
Biochimica et Biophysica Acta (BBA) - Lipids and Lipid Metabolism, 1984An abnormal acidic phospholipid was found in high concentration in kidney and brain, and also in other organs of rats exposed to ethanol by i.p. injection or by a liquid diet. The compound could be identified as phosphatidylethanol. Phosphatidylethanol is probably formed in cell membranes by a phospholipase D-catalyzed transphosphatidylation reaction.

[Formation of phosphatidylethanol in alcoholic intoxication].
Voprosy meditsinskoi khimii, 1988Phospholipid composition was studied in membranes of various tissues as well as of mitochondria and erythrocytes of rats with alcohol intoxication. A new phospholipid--phosphatidyl ethanol, which was absent in tissues of control animals, was detected in liver, heart, kidney, small intestine tissues as well as in erythrocytes and liver mitochondria of ...

Inter Individual Variation and Factors Regulating the Formation of Phosphatidylethanol
Alcoholism: Clinical and Experimental Research, 2019BackgroundAlcohol use disorders are a major but often unrecognized health problem. Alcohol markers can therefore be of great value for diagnosis, follow‐up, and treatment evaluation. Phosphatidylethanol in blood (B‐PEth) is an alcohol biomarker with higher clinical sensitivity and specificity than commonly used alcohol markers but has shown a ...

Phosphatidylethanol; clinical significance and biochemical basis
2001As shown in figure 1, ethanol can be metabolized by oxidative and nonoxidative pathways (Lieber, 1995). In the oxidative pathway, ethanol is converted to acetal-dehyde through the action of alcohol dehydrogenases, the microsomal ethanol-oxidizing system, or catalase.

Influence of Ethanol and Phosphatidylethanol on the Activity of Pancreatic Phospholipase A2
Biochemistry (Moscow), 2001Hydrolysis of dioleoylphosphatidylethanol (DOPEt) and dioleoylphosphatidylcholine (DOPC) catalyzed by phospholipase A2 (PLA2) from porcine pancreas has been studied in single-component and binary liposomes in the absence and in the presence of ethanol. DOPEt (an anionic phospholipid) was found to increase the rate of hydrolysis of zwitterionic DOPC in ...

Improving screening for alcohol consumption during pregnancy with phosphatidylethanol
Reproductive Toxicology, 2017The study objective was to compare rates of alcohol use between urine ethanol testing and self- reporting (Method: 1) and Phosphatidylethanol (PEth) dried blood spot testing and self-reporting (Method: 2).This was a prospective observational study in an obstetric clinic with universal alcohol screening.Method: 1 identified 11 patients with alcohol use (
