Evolution of PIKK family kinase inhibitors: A new age cancer therapeutics

open access: yesFrontiers in Bioscience-Landmark, 2020
Phosphatidylinositol-3 kinase-related kinases (PIKKs) belong to a family of atypical serine/threonine kinases in humans. They actively participate in a diverse set of cellular functions such as meiotic, V(D)J recombination, chromosome maintenance, DNA ...

A Requirement for Phosphatidylinositol 3-Kinase in Pseudopod Extension [PDF]

open access: yesJournal of Biological Chemistry, 1999
Phagocytosis requires actin assembly and pseudopod extension, two cellular events that coincide spatially and temporally. The signal transduction events underlying both processes may be distinct. We tested whether phagocytic signaling resembles that of growth factor receptors, which induce actin polymerization via activation of phosphatidylinositol 3 ...

mTORC2 Phosphorylation of GSDME‐N Drives Cullin4B‐Mediated Proteasomal Degradation to Suppress Pyroptosis and Confer Radioresistance in Small Cell Lung Cancer

open access: yesAdvanced Science, EarlyView.
Radioresistance severely limits the efficacy of therapies for small cell lung cancer (SCLC). This study reveals a novel mechanism of resistance driven by the active suppression of pyroptosis. Specifically, the mTORC2 complex directly phosphorylates GSDME‐N and promotes its CUL4B‐mediated ubiquitination and proteasomal degradation.

The role of myotubularin-related lipid phosphatases in cancer pathogenesis

open access: yesExperimental and Molecular Pathology
Myotubularin-related proteins (MTMRs) comprise a family of lipid phosphatases using phosphatidylinositol 3-phosphate and phosphatidylinositol 3,5-bisphosphate as their substrate.

Preparation, Component Identification, and Antioxidant Activity of Anthocyanins From Punica granatum “Tunisia”

open access: yesAgriFood: Journal of Agricultural Products for Food, EarlyView.
The preparation for Tunisian pomegranate anthocyanin extract. The molecular mechanisms underlying the antioxidant effects of these compounds were investigated using network pharmacology and molecular docking. Antioxidant activity was evaluated through DPPH, ABTS, and FRAP assays, whereas the effects on oxidative stress and inflammation were examined in

Long‐Term Follow Up of Two Patients With Variants in the Cluster 1031‐1159 of TRRAP Gene: Expanding the Phenotype of Developmental Delay With or Without Dysmorphic Facies and Autism

open access: yesAmerican Journal of Medical Genetics Part A, EarlyView.
ABSTRACT The transformation/transcription domain‐associated protein (TRRAP) gene encodes a large multidomain protein, a member of the phosphatidylinositol 3‐kinase‐related kinase (PIKK) family. TRRAP is a component of the histone acetyltransferase (HAT) complex, and it plays an important role in gene transcription, DNA repair, and cell‐cycle regulation.

CHF6523 data suggest that the phosphoinositide 3-kinase delta isoform is not a suitable target for the management of COPD

open access: yesRespiratory Research
Background Chronic obstructive pulmonary disease (COPD) is a chronic inflammatory condition. Given patients with COPD continue to experience exacerbations despite the availability of effective therapies, anti-inflammatory treatments targeting novel ...
