[26] Enzymatic synthesis of phytoene

1985
Publisher Summary This chapter focuses on enzymatic synthesis of phytoene. Enzyme systems in the chromoplasts of tomato fruits and in extracts of spinach leaves convert isopentenyl pyrophosphate to phytoene by the reactions are discussed in the chapter.

The purification of phytoene dehydrogenase from Phycomyces blakesleeanus

Biochimica et Biophysica Acta (BBA) - Lipids and Lipid Metabolism, 1994
The carotenogenic enzyme phytoene dehydrogenase has been purified from the C9carR21(-) (lycopene-accumulating) mutant of the filamentous fungus Phycomyces blakesleeanus. Solubilization of the membrane-bound enzyme with 1% Tween-60 was followed by a 250-fold purification to homogeneity using polyethylene glycol precipitation, CM-Sepharose, gel ...

The occurrence of phytoene in various plant materials

Archives of Biochemistry and Biophysics, 1953
Abstract A method of assay for phytoene in plant materials is presented which involves isooctane-ethanol extraction, adsorption of the interfering substances on a magnesia-Super Cel column, and absorptimetric determination of phytoene at 286 mμ. Twenty plant materials were analyzed for phytoene by this method.

Accumulation of phytoene, a colorless carotenoid by inhibition of phytoene desaturase (PDS) gene in Dunaliella salina V-101

Bioresource Technology, 2017
The aim of this work was to study the accumulation of phytoene in Dunaliella salina V-101 by down-regulating its phytoene desaturase (PDS) gene expression using RNA interference and Antisense technology. RNAi and antisense constructs were introduced into the Dunaliella cells by Agrobacterium-mediated transformation.

Production of phytoene, a carotenoid, and induction of connexin 26 in transgenic mice carrying the phytoene synthase gene crtB

Biochemical and Biophysical Research Communications, 2004
Carotenoids have been recognized as chemopreventive agents against human diseases, such as cancer and cardiovascular disease. Mammalians utilize carotenoids supplied from their food since they are unable to perform the de novo synthesis of carotenoids.

The stereochemistry of phytoene biosynthesis

Proceedings of the Royal Society of London. Series B. Biological Sciences, 1966
Experiments with carrot root preparations have demonstrated that all the eliminations of hydrogen at C-4 of mevalonic acid during its conversion into phytoene have the same stereospecificity; the 4- R hydrogen is retained and the 4 S hydrogen is eliminated. The same stereospecific elimination

Genetic control of phytoene dehydrogenation in phycomyces

Plant Science Letters, 1974
Abstract Strains of Phycomyces blakesleeanus carrying mutations in gene carB accumulate large amounts of phytoene. A leaky carB mutant, isolated from a red, lycopene-producing strain, accumulates decreasing amounts of phytoene, phytofluene, ζ-carotene, neurosporene, and lycopene.

[Advances in phytoene dehydrogenase - A review].

Wei sheng wu xue bao = Acta microbiologica Sinica, 2018
Carotenoids, as a group of over 700 valuable unsaturated terpene compounds classified as carotene and xanthophyll family, are endowed with powerful nutritional value. Phytoene dehydrogenase is the key rate-limiting enzyme in carotenoids biosynthesis pathway, involved in catalyzing the conversion from colorless hydrocarbon phytoene to other pigmented ...

Phytoene Desaturase: Genes, Enzymes and Phylogenetic Aspects

Journal of Plant Physiology, 1994
Summary Recent work on phytoene desaturases and also on ζ-carotene desaturase is presented and discussed. This involves functional complementation of phytoene desaturation by heterologous expression of different genes coding for this enzyme originating from Rhodobacter capsulatus, Erwinia uredovora, and Synechococcus and the enzymatic ...
