Evaluating Sampling Designs to Survey Fish Diversity in Lakes From Northern Temperate Zones

We demonstrate that, under certain circumstances, stratified sampling requires fewer samples than random sampling to detect 90% and 95% of species in 12 temperate freshwater lakes using the 12S MiFish metabarcoding. We also demonstrate that a sequencing depth of 50K reads per sample is adequate for a reliable portrayal of species richness.
