Piezo1 channel exaggerates ferroptosis of nucleus pulposus cells by mediating mechanical stress-induced iron influx

open access: yesBone Research
To date, several molecules have been found to facilitate iron influx, while the types of iron influx channels remain to be elucidated. Here, Piezo1 channel was identified as a key iron transporter in response to mechanical stress.

Stimulation of 3D osteogenesis by mesenchymal stem cells using a nanovibrational bioreactor [PDF]

open access: yes, 2017
Bone grafts are one of the most commonly transplanted tissues. However, autologous grafts are in short supply, and can be associated with pain and donor-site morbidity.

Sphingomyelinase Disables Inactivation in Endogenous PIEZO1 Channels [PDF]

open access: yesCell Reports, 2020
Endogenous PIEZO1 channels of native endothelium lack the hallmark inactivation often seen when these channels are overexpressed in cell lines. Because prior work showed that the force of shear stress activates sphingomyelinase in endothelium, we considered if sphingomyelinase is relevant to endogenous PIEZO1. Patch clamping was used to quantify PIEZO1-

Ultrasound Modulation of Visual Circuits in Mice Independent of Auditory Confound

open access: yesAdvanced Science, EarlyView.
In this study, Qiu et al. found that low‐intensity ultrasound can directly activate sparse ultrasound‐sensitive neurons (UNs) in the primary visual cortex (V1) of deafened mice. The proportion of these sparse UNs is pressure‐dependent. Furthermore, ultrasound modulates visual circuitry with distinct excitatory and inhibitory effects.

Single mechanosensitive and ca(2+)-sensitive channel currents recorded from mouse and human embryonic stem cells [PDF]

open access: yes, 2015
Cell-attached and inside-out patch clamp recording was used to compare the functional expression of membrane ion channels in mouse and human embryonic stem cells (ESCs). Both ESCs express mechanosensitive Ca(2+) permeant cation channels (MscCa) and large

Liver Stiffness Rises Early in MASLD and Drives Inflammation, Lipid Dysmetabolism, and Fibrosis via Piezo1–YAP Mechanotransduction

open access: yesAdvanced Science, EarlyView.
Progressive stiffening of the hepatic extracellular matrix and free fatty acid overload activate the mechanosensor Piezo1 in steatotic hepatocytes, inducing calcium influx and nuclear translocation of Yes‐associated protein. YAP‐dependent nuclear signaling further drives inflammatory and fibrotic gene programs, lipid metabolic reprogramming, and ...

Reversal of Stress‐Induced PIEZO1 Elevation with Mechanically Adapted Epicardial Patch for Myocardial Infarction Treatment

open access: yesAdvanced Science
Elevated expression of the mechanosensitive ion channel PIEZO1 in response to abnormal mechanical stimuli is implicated in many diseases, including myocardial infarction (MI).

The role of MscL amphipathic N terminus indicates a blueprint for bilayer-mediated gating of mechanosensitive channels [PDF]

open access: yes, 2016
The bacterial mechanosensitive channel MscL gates in response to membrane tension as a result of mechanical force transmitted directly to the channel from the lipid bilayer.

Functional Heteromeric PIEZO1 Ion Channels [PDF]

open access: yesBiophysical Journal, 2016
Eukaryotic PIEZO1 ion channels are cation selective and open with membrane tension. The channel is further characterized by voltage dependent inactivation and a conductance of ∼45 pS. We mutated the glutamic acid at position 2117 to an aspartic acid and this increased channel conductance to 72 pS.
