A Protein‐Centric Strategy Coupled with Match‐Between‐Run Glycoproteomics Enables Discovery of Robust Site‐Specific Glycan Biomarkers for Hepatocellular Carcinoma

In this study, a highly robust N‐glycoproteomics (HRN) platform with a newly developed match‐between‐run scheme is used for biomarker discovery. This study proposes a protein‐centric strategy to prioritize proteins susceptible to aberrant glycosylation.

Immunohistochemical study for PIVKA-II in hepatocellular carcinoma.

吸引肝生検で診断された肝細胞癌(HCC)患者70例(A群:腫瘍径2cm以下23例,B群:径2~5cm 13例,C群:径5cm以上および多結節34例)の肝癌組織を抗PIVKA-II抗体を用いた酵素抗体間接法で染色し光顕・電顕的に観察した.HCC 70例のうち組織PIVKA-II染色陽性は29例(41.4%)であった.細胞異型度別の陽性率には有意差を認めなかったが,中等度異型度群(Ed-II)で強く染色される傾向があった.腫瘍径別の陽性率はA群39.1%, B群38.5%, C群44.1%であり,A群での血中PIVKA-II陽性例は1例のみであった.また,腫瘍切除後に血中PIVKA-IIの正常化した症例が認められたことや電顕的に肝癌細胞の粗面小胞体にPIVKA-IIに対する反応産物が認められたことから ...
