Discovery of allostery in PKA signaling
cAMP-dependent protein kinase (PKA) was the second protein kinase to be discovered and the PKA catalytic (C) subunit serves as a prototype for the large protein kinase superfamily that contains over 500 gene products. The protein kinases regulate much of biology in eukaryotic cells and they are now also a major therapeutic target.

PKA inhibits WNT signalling in adrenal cortex zonation and prevents malignant tumour development
Adrenal cortex physiology relies on functional zonation, essential for production of aldosterone by outer zona glomerulosa (ZG) and glucocorticoids by inner zona fasciculata (ZF).

A Functionally Conserved Enhancer is Critical for PGC1A Expression and Thermogenesis in Brown Fat
An evolutionarily conserved enhancer controls PGC1A expression and thermogenic activation in brown adipose tissue. Integrative multi‐omics analyses identify two BAT‐specific enhancers, with PGC1A‐En1 functioning as the dominant regulatory element that maintains thermogenic capacity across species.

In our previous study, anti-apoptotic effects of GABAC-receptor stimulation was suppressed by inhibitors of cAMP-dependent protein kinase (PKA), implying GABACreceptor–mediated PKA activation. The present study showed that GABAC-receptor stimulation with

Interleukin 2 transcription factors as molecular targets of cAMP inhibition: delayed inhibition kinetics and combinatorial transcription roles [PDF]
Elevation of cAMP can cause gene-specific inhibition of interleukin 2 (IL-2) expression. To investigate the mechanism of this effect, we have combined electrophoretic mobility shift assays and in vivo genomic footprinting to assess both the availability ...

Combating VEGFA‐siRNA‐Induced Metabolic Reprogramming via Glucose Utilization Deprivation
An ionizable lipid nanoparticle co‐delivers VEGFA siRNA and glucose oxidase to tumors to counteract VEGFA knockdown‐induced, glutamine‐driven metabolic compensation. Enzymatic glucose consumption reverses the adaptive pathways and restores metabolism toward baseline, thereby sensitizing tumors to therapy.

G3BP1 Succinylation at K413 is Critical for Cardiac Function by Modulating PI3K‐AKT‐mTOR Signal Axis
Schematic illustrating the impact of G3BP1 succinylation at K413 on cardiac function. In the healthy human heart, G3BP1 succinylation maintains homeostatic mTOR signaling. In patients with dilated cardiomyopathy (DCM) and heart failure (HF), G3BP1 de‐succinylation induces RagA expression and disrupts the binding of the TSC1/2 complex, leading to the ...
