Strategy to develop and validate digital droplet PCR methods for global antimicrobial resistance wastewater surveillance

open access: yesWater Environment Research, Volume 96, Issue 11, November 2024.
Workflow for the development and validation of novel ddPCR methods for a harmonized and global AMR monitoring in wastewater. Abstract According to World Health Organization (WHO), antimicrobial resistance (AMR) is currently one of the world's top 10 health threats, causing infections to become difficult or impossible to treat, increasing the risk of ...

New insights into the mechanism of action of pyrazinamide, implications for susceptibility testing, and future regimens*

open access: yesInternational Journal of Mycobacteriology, 2016
Pyrazinamide (PZA) is included in the 2016 World Health Organization multidrug-resistant tuberculosis treatment guidelines and is a key component of most ongoing clinical trials investigating novel antibiotic combinations.

Insight into novel clinical mutants of RpsA-S324F, E325K, and G341R of Mycobacterium tuberculosis associated with pyrazinamide resistance

open access: yesComputational and Structural Biotechnology Journal, 2018
Pyrazinamide (PZA) is an important component of first-line anti-tuberculosis drugs which is converted into active form, pyrazinoic acid (POA), by Mycobacterium tuberculosis (MTB) pncA gene encoded, pyrazinamidase (PZase).

The Study of PncA Gene Using PCR-RFLP and Allele-Specific PCR Methods in Distinguishing Mycobacterium Bovis from Mycobacterium Tuberculosis

open access: greenمجله دانشکده پزشکی اصفهان, 2011
Background: Bovine tuberculosis is a chronic bacterial disease of cattle that occasionally affects human. A unique mutation at position 169 (C→G) in Mycobacterium bovis differentiates it from Mycobacterium tuberculosis. For the first time, PCR-RFLP method was used in this study to separate the two organisms.

[Investigation of pncA, rpsA and panD Gene Mutations Associated with Resistance in Pyrazinamide-Resistant Mycobacterium tuberculosis Isolates and Spoligotyping].

open access: yesMikrobiyoloji bulteni, 2022
Pyrazinamide (PZA) is one of the first-line anti-tuberculous drugs used in the treatment of tuberculosis (TB). Considering the ability of PZA to shorten the treatment period from 9-12 months to six months by eliminating persistent bacilli, it appears to be an important cornerstone of TB therapy.

Estimation of pyrazinamidase activity using a cell-free In vitro synthesis of pnca and its association with pyrazinamide susceptibility in Mycobacterium tuberculosis

open access: yesInternational Journal of Mycobacteriology, 2018
Background: The main mechanism of resistance to PZA in Mycobacterium tuberculosis relies on mutations on its pyrazinamidase/nicotinamidase. Recently, a rapid colorimetric test relying on the PCR-based in vitro-synthesized-PZase assay has been reported ...

Molecular analysis of pyrazinamide resistance in Mycobacterium tuberculosis in Vietnam highlights the high rate of pyrazinamide resistance-associated mutations in clinical isolatesPncA mutations and PZA resistance in M. tuberculosis in Vietnam

open access: yesEmerging Microbes and Infections, 2017
Pyrazinamide (PZA) is a key antibiotic in current anti-tuberculosis regimens. Although the WHO has stressed the urgent need to obtain data on PZA resistance, in high tuberculosis burden countries, little is known about the level of PZA resistance, the ...
