Summary Specific antibody against poly(ADP-Rib) was produced in a rabbit by injecting poly(ADP-Rib) mixed with methylated bovine serum albumin. Under standardized conditions, 1 mg of purified anti-poly(ADP-Rib) antibody combined with 400 pmoles (4 μg) of poly(ADP-Rib) and was retained on a millipore filter.

Poly(adenosine 5'-diphosphate ribose) [poly(ADP-ribose]) is spontaneously ADP-ribosylated when it is incubated with nicotinamide adenine dinucleotide, especially in 0.5 M NaCl and at an alkaline pH. The ADP-ribose residues are monomeric and are attached to the middle of polymer chains.

The chromatin-associated enzyme poly(ADP-Rib) polymerase catalyzes the posttranslational modification of histones. Antibody to poly(ADP-Rib) has been coupled to Sepharose, and the resultant immunoadsorbent was used to fractionate, specifically, histone H1 subpopulations undergoing this nuclear protein modification.
