Characterization of polyhydroxyalkanoate synthases from the marine bacterium Neptunomonas concharum JCM17730

Journal of Biotechnology, 2020
Neptunomonas concharum JCM17730 was isolated from an ark clam sample and characterized as a mesophilic bacterium. The genome of N. concharum JCM17730 contains thirteen genes related to polyhydroxyalkanoates (PHA) metabolism. Three PHA synthase encoding genes were identified, and phylogenetic analysis of enzyme sequences suggested the presence of two ...

Natural and engineered polyhydroxyalkanoate (PHA) synthase: key enzyme in biopolyester production

Applied Microbiology and Biotechnology, 2017
With the finite supply of petroleum and increasing concern with environmental issues associated with their harvest and processing, the development of more eco-friendly, sustainable alternative biopolymers that can effectively fill the role of petro-polymers has become a major focus.

Thiolysis of Poly(3-hydroxybutyrate) with Polyhydroxyalkanoate Synthase from Ralstonia eutropha

The Journal of Biochemistry, 2006
Poly(3-hydroxybutyrate) (PHB) is synthesized from 3-hydroxybutyryl-CoA by polyhydroxyalkanoate synthase and hydrolyzed by PHB depolymerase. In this study, we focused on the reverse reaction of polyhydroxyalkanoate synthase, and propose the possibility that PHB can be degraded through a novel process, that is thiolysis of PHB with CoASH ...

Regulation of polyhydroxyalkanoate synthases (phaC1 and phaC2) gene expression in Pseudomonas corrugata

Applied Microbiology and Biotechnology, 2006
In this study we examined polyhydroxyalkanoate (PHA) synthases phaC1 and phaC2 gene expression in two strains of Pseudomonas corrugata (Pc) grown in a minimum mineral medium with related (oleic acid and octanoate) or unrelated (glucose) carbon sources.

Characterization of the Polyhydroxyalkanoate Synthase Gene Locus of Rhodobacter Sphaeroides

Biotechnology Letters, 1993
The Rhodobacter sphaeroides polyhydroxyalkanoate synthase structural gene (phaCRs) was identified as a 1,806-bp open reading frame. It encoded a PHA synthase of 601 amino acids with a relative molecular mass of 66,742, which exhibited 25.9 to 39.0 % amino acid identity with PHA synthases from other bacteria.

Increased synthesis of poly(3-hydroxydodecanoate) by random mutagenesis of polyhydroxyalkanoate synthase

Applied Microbiology and Biotechnology, 2018
Poly(3-hydroxydodecanoate) [P(3HDD)], a medium-chain-length polyhydroxyalkanoate (PHA), is expected to be used as a novel type of bioplastic characterized by a soft and transparent nature. In this study, to achieve a high yield of P(3HDD), PHA synthase was modified through random mutagenesis of a region of the PHA synthase 1 gene from Pseudomonas ...

Cloning and characterization of the Methylobacterium extorquens polyhydroxyalkanoic-acid-synthase structural gene

Applied Microbiology and Biotechnology, 1993
A cosmid gene bank of partially EcoRI-digested genomic DNA from Methylobacterium extorquens IBT no. 6 was screened for DNA fragments restoring polyhydroxyalkanoic-acid (PHA) accumulation in the PHA-negative mutant Alkaligenes eutrophus H16 PHB-4. The M.

Cloning, characterization and comparison of the Pseudomonas mendocina polyhydroxyalkanoate synthases PhaC1 and PhaC2

Applied Microbiology and Biotechnology, 2002
This study describes a comparison of the polyhydroxyalkanoate (PHA) synthases PhaC1 and PhaC2 of Pseudomonas mendocina. The P mendocina pha gene locus, encoding two PHA synthase genes [phaC1Pm and phaC2pm flanking a PHA depolymerase gene (phaZ)], was cloned, and the nucleotide sequences of phaC1Pm (1,677 bp), phaZ (1,034 bp), and phaC2pm (1,680 bp ...

Effect of increased PHA synthase activity on polyhydroxyalkanoates biosynthesis in Synechocystis sp. PCC6803

International Journal of Biological Macromolecules, 2002
Polyhydroxyalkanoate (PHA) synthase activity in Synechocystis sp. PCC6803 was increased two-fold by introducing the PHA biosynthetic genes of Ralstonia eutropha. The resulting recombinant Synechocystis sp. PCC6803 strain was subjected to conditions that favor PHA accumulation and the effects of various carbon sources were studied. In addition, the fine

Insights into the surface topology of polyhydroxyalkanoate synthase: self-assembly of functionalized inclusions

Applied Microbiology and Biotechnology, 2015
The polyhydroxyalkanoate (PHA) synthase catalyzes the synthesis of PHA and remains attached to the hydrophobic PHA inclusions it creates. Although this feature is actively exploited to generate functionalized biobeads via protein engineering, little is known about the structure of the PHA synthase.
