Investigation of the Involvement of NLRP3 Inflammasome Activation in the Proinflammatory Response to Clozapine [PDF]

open access: yes, 2022
Idiosyncratic drug reactions (IDRs) represent a major concern in the development of new drugs as they are rarely identified in clinical trials and can be severe and life-threatening in nature, putting a given drug’s success at risk.

Virus Binding to a Plasma Membrane Receptor Triggers Interleukin-1α-Mediated Proinflammatory Macrophage Response In Vivo [PDF]

open access: yes, 2009
SummaryThe recognition of viral components by host pattern-recognition receptors triggers the induction of the antiviral innate immune response. Toll-like receptor 9 (TLR9) and NLRP3 inflammasome were shown to be the principal specific sensors of viral ...

TOLLIP Inhibits Psoriasis Progression via Suppressing PKM2‐Mediated Glycolysis in Keratinocytes

open access: yesAdvanced Science, EarlyView.
In this study, we identify TOLLIP as a critical regulator of psoriasis pathogenesis through its modulation of glycolytic metabolism. Our findings establish the TOLLIP‐PKM2‐glycolysis axis as a key mechanism linking metabolic reprogramming to psoriasis pathogenesis, and propose TOLLIP as a promising therapeutic target.

Lactiplantibacillus Plantarum GUANKE Alleviates Allergic Asthma by Restricting Monocyte‐to‐DC Differentiation Via Microbiota‐Associated IAA

open access: yesAdvanced Science, EarlyView.
Lactiplantibacillus plantarum GUANKE remodels the indigenous gut microbiota and is associated with microbiota‐dependent restoration of circulating IAA. IAA treatment attenuates JAK3‐STAT3 and Jag1‐Notch4‐Hey1 signaling and reduces CX3CR1+Ly6C− monocytic precursor generation and pulmonary CD11b+ DC accumulation, together with reduced Th2 inflammation ...

RIPK4 activates an IRF6-mediated proinflammatory cytokine response in keratinocytes [PDF]

open access: yes, 2016
Keratinocytes of the oral mucosa and epidermis play key roles in host defense. In addition to functioning as a physical barrier, they also produce cytokines to elicit inflammation in response to infection or injury.

Celastrol‐Loaded Conductive Hydrogel Mitigates Myocardial Ischemia‐Reperfusion Injury and Restores Electrophysiological Function

open access: yesAdvanced Science, EarlyView.
Schematic illustration of the Pluronic F127 diacrylate/gelatin methacryloyl/poly(3,4‐ethylenedioxythiophene):poly(styrenesulfonate)/celastrol hydrogel (FGPC) for localized myocardial ischemia–reperfusion injury treatment. FGPC combines sustained celastrol delivery with PEDOT:PSS‐mediated conductive support to attenuate acute oxidative/inflammatory ...

Engineering Microbial Particles for Next‐Generation Biomedical Platforms

open access: yesAdvanced Science, EarlyView.
Microbe‐derived particles (MDPs), which include extracellular vesicles, outer membrane vesicles, inclusion bodies, polysaccharide particles, and virus‐like particles, represent a rapidly expanding category of bioinspired nanomaterials. With their natural origin, intrinsic biocompatibility, and highly programmable functionality, MDPs serve as a ...

MAM‐Localized MANF Counteracts Microinflammatory Stress to Attenuate Mitochondrial Dysfunction and Cataractogenesis in High Myopia

open access: yesAdvanced Science, EarlyView.
Chronic microinflammation in high myopia suppresses MANF in lens epithelial cells. MANF normally resides within MAMs and promotes ubiquitin‐mediated degradation of the ER Ca2+ pump SERCA2. MANF loss causes pathological SERCA2 accumulation, MAM hyperassembly, and disrupted ER‐to‐mitochondria Ca2+ transfer, leading to mitochondrial failure, oxidative ...
