MeCP2 Lactylation Protects against Ischemic Brain Injury by Transcriptionally Regulating Neuronal Apoptosis

MeCP2 lactylation protects against ischemic stroke by reducing brain infarct volume and improving neurological outcomes. Lactylation at K210/K249 exerts neuroprotective effects by repressing the transcription of apoptosis‐associated genes, including Pdcd4 and Pla2g6. HDAC3 and p300 serve as key regulatory enzymes for MeCP2 lactylation following stroke.

Diallyl trisulfides (DATs) selectively induce cuproptosis in hepatic stellate cells (HSCs) by targeting Ras‐related protein Rab‐18 (RAB18) and regulating lipophagy. DATs promote RAB18 phase separation, enhance mitochondrial‐associated membrane structures (MAMs) formation, and increase succinylation of dihydrolipoamide dehydrogenase (DLD) at K320.

RAB3B Dictates mTORC1/S6 Signaling in Chordoma and Predicts Response to mTORC1‐Targeted Therapy

RAB3B is unveiled as a prominent oncogenic regulator in chordoma, which can block the DUSP12‐mediated dephosphorylation of p‐S6 (S235/236). The combination of RAB3B and p‐S6 indicates a good prognostic value and predicts mTORC1 inhibitors response for chordoma patients.

Development of MDM2‐Targeting PROTAC for Advancing Bone Regeneration

This work presents a creative pipeline for developing MDM2‐targeting PROTACs (MDM2‐PROTACs) for application in bone regeneration. The developed PROTACs (CL144 and CL174) are validated for their degradation efficiency and osteogenic effects in human BMSCs.
