Elucidation of interface interactions between a dehydratase domain and an acyl carrier protein in cremimycin polyketide synthase

open access: yesFEBS Letters, EarlyView.
In modular polyketide synthases, the dehydratase (DH) domain catalyzes the dehydration reaction of the β‐hydroxyacyl unit attached to the cognate acyl carrier protein (ACP) domain. However, it is unclear how DH interacts with ACP during the reaction. In this study, we identified DH–ACP interface residues, providing the first detailed insights into DH ...

The diverse dependence of galectin-1 and -8 on multivalency for the modulation of FGFR1 endocytosis

open access: yesCell Communication and Signaling
Fibroblast growth factor receptor 1 (FGFR1) is a N-glycosylated cell surface receptor tyrosine kinase, which upon recognition of specific extracellular ligands, fibroblast growth factors (FGFs), initiates an intracellular signaling.

Functional variation among LPMOs revealed by the inhibitory effects of cyanide and buffer ions

open access: yesFEBS Letters, EarlyView.
This study addresses the inhibition of lytic polysaccharide monooxygenases (LPMOs) by cyanide and explains how and why the magnitude of observed inhibitory effects depends on the way LPMO reactions are setup and on the type of LPMO. Enzymes known as lytic polysaccharide monooxygenases (LPMOs) are mono‐copper polysaccharide‐degrading peroxygenases that ...

Deep Generative Modeling for Protein Design [PDF]

open access: yesarXiv, 2021
Deep learning approaches have produced substantial breakthroughs in fields such as image classification and natural language processing and are making rapid inroads in the area of protein design. Many generative models of proteins have been developed that encompass all known protein sequences, model specific protein families, or extrapolate the ...

Ablation of LRP6 in alpha‐smooth muscle actin‐expressing cells abrogates lung inflammation and fibrosis upon bleomycin‐induced lung injury

open access: yesFEBS Letters, EarlyView.
Low‐density lipoprotein receptor‐related protein 6 (LRP6) is a key receptor for the Wnt antagonist Dickkopf1 (DKK1). DKK1 protein expression is induced in a bleomycin (BLM)‐induced lung injury model. We show that DKK1 induces proinflammatory and profibrotic genes in lung fibroblasts.

An Extended Reservoir of Class-D Beta-Lactamases in Non-Clinical Bacterial Strains

open access: yesMicrobiology Spectrum, 2022
Bacterial genes coding for antibiotic resistance represent a major issue in the fight against bacterial pathogens. Among those, genes encoding beta-lactamases target penicillin and related compounds such as carbapenems, which are critical for human ...

Characteristics of the Kelch domain containing (KLHDC) subfamily and relationships with diseases

open access: yesFEBS Letters, EarlyView.
The Kelch protein superfamily includes 63 members, with the KLHDC subfamily having 10 proteins. While their functions are not fully understood, recent advances in KLHDC2's structure and role in protein degradation have highlighted its potential for drug development, especially in PROTAC therapies.

Identification of a peptide antagonist of the FGF1–FGFR1 signaling axis by phage display selection

open access: yesFEBS Open Bio, 2019
Overexpression of fibroblast growth factor receptor 1 (FGFR1) is a common aberration in lung and breast cancers and has necessitated the design of drugs targeting FGFR1‐dependent downstream signaling and FGFR1 ligand binding.

Grabbing the Bull by Both Horns: Bovine Ultralong CDR-H3 Paratopes Enable Engineering of ‘Almost Natural’ Common Light Chain Bispecific Antibodies Suitable For Effector Cell Redirection

open access: yesFrontiers in Immunology, 2022
A subset of antibodies found in cattle comprises ultralong CDR-H3 regions of up to 70 amino acids. Interestingly, this type of immunoglobulin usually pairs with the single germline VL gene, V30 that is typically very conserved in sequence.

Insertion of the FeB cofactor in cNORs lacking metal inserting chaperones

open access: yesFEBS Letters, EarlyView.
Nitric oxide reductase is an enzyme found in the bacterial denitrification pathway. The NOR active site contains a non‐heme iron, often, but not always inserted with the assistance of chaperones. Here, we study the insertion of FeB in the subfamily of cNORs lacking chaperones and found a putative channel, conserved in the family, perhaps enabling the ...
