Nuclear protein import in permeabilized mammalian cells requires soluble cytoplasmic factors

open access: yesJournal of Cell Biology, 1990
We have developed an in vitro system involving digitonin-permeabilized vertebrate cells to study biochemical events in the transport of macromolecules across the nuclear envelope.

Cytochrome oxidase subunit VI of Trypanosoma brucei is imported without a cleaved presequence and is developmentally regulated at both RNA and protein levels [PDF]

open access: yes, 2001
Mitochondrial respiration in the African trypanosome undergoes dramatic developmental stage regulation. This requires co-ordinated control of components encoded by both the nuclear genome and the kinetoplast, the unusual mitochondrial genome of these ...

Elastomeric 3D‐Printed Microenvironments Enable Nanonewton Force Measurements in Healthy and Diseased Human Pluripotent Stem Cell‐Derived Neuroepithelial Cells

open access: yesAdvanced Materials, EarlyView.
We present elastomeric three‐dimensional (3D) microstructures fabricated via two‐photon polymerization (2PP) and post‐processed through wet etching, for quantifying nanonewton (nN)‐scale forces applied by healthy and diseased neural cells. The mechanically characterized free‐standing beam architectures enable measurement of traction forces of ...

Border Control: Selectivity of Chloroplast Protein Import and Regulation at The Toc-Complex

open access: yesFrontiers in Plant Science, 2014
Plants have evolved complex and sophisticated molecular mechanisms to regulate their development and adapt to their surrounding environment. Particularly the development of their specific organelles, chloroplasts and other plastid-types, is finely tuned ...

Translocation and insertion of precursor proteins into isolated outer membranes of mitochondria [PDF]

open access: yes, 1993
Nuclear-encoded proteins destined for mitochondria must cross the outer or both outer and inner membranes to reach their final sub- mitochondrial locations. While the inner membrane can translocate preproteins by itself, it is not known whether the outer

Nuclear Pore Mechanotransduction in Oncology: A Structural Axis of Vulnerability for Targeted Intervention

open access: yesAdvanced Materials, EarlyView.
Cancer‐associated NPC remodeling creates a high‐flux, low‐stringency nuclear state that supports malignant adaptation but increases mechanical fragility. Targeting the FG‐barrier or NPC scaffold may drive mechanostat failure, envelope rupture, DNA damage, and loss of nuclear integrity.

Bottom‐Up Engineering of a Human Neuromuscular System for Modeling Activity‐Induced Remodeling, Metabolic Stress, and Endothelial‐Modulated Excitability

open access: yesAdvanced Materials, EarlyView.
Using a bottom‐up approach, we engineered a human neuromuscular microchip to investigate distinct adaptive responses to neural stimulation and metabolic stress. Further integration of endothelial cells revealed their critical role in modulating neuromuscular excitability, establishing a comprehensive neurovascular‐muscular model.

Lab‐in‐a‐Tube System Integrating Hierarchically Wrinkled Electrodes and DNAzyme‐Functionalized Beads for Diagnosis of Clostridioides difficile Infection

open access: yesAdvanced Materials, EarlyView.
The point‐of‐care diagnosis of Clostridioides difficile infection remains challenging due to reliance on slow culture and amplification‐based methods. A lab‐in‐a‐tube electrochemical biosensor integrating DNAzymes, antifouling magnetic beads, and a hierarchically structured flow cell enables amplification‐ and culture‐free detection directly in stool ...

Surface Functionalization Modulates Collective Cell Behavior Near Topological Defects Induced by Topographical Patterning

open access: yesAdvanced Materials Interfaces, EarlyView.
Strongly adhered cells align with concentric circular ridges and exhibit predominantly tangential motion, whereas weakly adhered cells fail to follow topographical guidance and display enhanced radial inward motion. ABSTRACT Living cells establish long‐range orientational order through collective alignment, giving rise to topological defects whose ...

Inhibition of IGFBP4 in Granulosa Cells Improves Reproductive Performance and Maintains Fertility With Age via YAP Signaling

open access: yesAdvanced Science, EarlyView.
IGFBP4 is upregulated in granulosa cells of aged ovaries across monkeys, mice, and humans. It inhibits YAP signaling, thereby suppressing cell proliferation and contributing to follicular dysfunction. Deletion of Igfbp4 in granulosa cells enhances ovulatory output, improves hormone profiles, and reproductive performance in aged female mice, suggesting ...
