Pyrimethamine was one of the first of many 2,4-diaminopyrimidines to be synthesized and tested for antimicrobial activity. Its chemical formula is CHClN, and its structure is shown in Figure 93.1. The main value of pyrimethamine has been its antiprotozoal activity, especially against malaria and Toxoplasma, for which it is used in combination regimens (

Field studies were conducted in Kisumu, Kenya, to assess the susceptibility of local strains of Plasmodium falciparum to pyrimethamine alone (by a standard 7-day in-vivo test and a 48 h in-vitro field test) and to sulfadoxine-pyrimethamine (by a 7-day in-vivo test).

Treatment of vivax malaria with sulfadoxine-pyrimethamine and with pyrimethamine alone
Transactions of the Royal Society of Tropical Medicine and Hygiene, 1979The effect of pyrimethamine and the combination of pyrimethamine-sulfadoxine (Fansidar) upon the termination of the acute attack of vivax malaria was studied in Thailand. Pyrimethamine was found to be ineffective, providing clearance of parasitaemia in only two of six patients by the end of seven days following treatment.

Abstract Pyrimethamine, an amyl analog of 2,4-diaminopyrimidine, is used in treatment and prophylaxis of toxoplasmosis and malaria; it is usually employed in combination with sulfon amides or sulfones. Pyrimethamine inhibits difolatereduc tase of microorganisms and prevents nuclear division dur ing mitosis; this inhibition occurs at ...
