Cysteine catabolism and the serine biosynthesis pathway support pyruvate production during pyruvate kinase knockdown in pancreatic cancer cells

open access: yesCancer & Metabolism, 2019
Background Pancreatic ductal adenocarcinoma (PDAC) is an aggressive cancer with limited treatment options. Pyruvate kinase, especially the M2 isoform (PKM2), is highly expressed in PDAC cells, but its role in pancreatic cancer remains controversial.

Role of pyruvate kinase M20-regulated glycolysis in inflammatory activation of macrophages in diabetic nephropathy

open access: yesDi-san junyi daxue xuebao, 2020
Objective To explore the role of pyruvate kinase M20 (PKM2)-regulated glycolysis on nod-like receptor protein 3 (NLRP3)- and absentin melanoma 2 (AIM2)-mediated inflammatory activation of macrophages in diabetic nephropathy (DN).

Glucose deprivation activates a metabolic and signaling amplification loop leading to cell death. [PDF]

open access: yes, 2012
The altered metabolism of cancer can render cells dependent on the availability of metabolic substrates for viability. Investigating the signaling mechanisms underlying cell death in cells dependent upon glucose for survival, we demonstrate that glucose ...

Changes in adenosine 5'-monophosphate-activated protein kinase as a mechanism of visceral obesity in Cushing's syndrome [PDF]

open access: yes, 2008
OBJECTIVE: Features of the metabolic syndrome such as central obesity with insulin resistance and dyslipidemia are typical signs of Cushing's syndrome and common side effects of prolonged glucocorticoid treatment.

Prmt6 Deficiency or Inhibition Restores Microglial Homeostasis and Promotes Scar‐Limited Repair in Adult Spinal Cord Injury

open access: yesAdvanced Science, EarlyView.
After spinal cord injury, adult microglia remain persistently activated with chronic PRMT6 (protein arginine methyltransferase 6) upregulation. Prmt6 deficiency or inhibition reestablishes microglial homeostasis and promotes a scar‐limited repairment, enhancing axonal regrowth.

HBV-Induced Pyruvate Increases Lactylation of Pyruvate Kinase M2 (PKM2) at K206 to Promote Liver Fibrosis

open access: yesPathogens
We previously demonstrated that HBV promotes liver fibrosis through the enhanced production of pyruvate. Pyruvate kinase M2 (PKM2), a key enzyme in pyruvate metabolism, plays an important role in liver fibrogenesis. Recently, lactylation of PKM2 has been

Loss of pyruvate kinase M2 limits growth and triggers innate immune signaling in endothelial cells

open access: yesNature Communications, 2018
The glycolytic enzyme pyruvate kinase M2 (PKM2) is required for nucleotide synthesis and cell proliferation. Using gene expression and metabolomics analyses, the authors here show that PKM2 regulates methionine metabolism and DNA methylation in ...

Pyruvate kinase M2 activators promote tetramer formation and suppress tumorigenesis [PDF]

open access: yes, 2014
Cancer cells engage in a metabolic program to enhance biosynthesis and support cell proliferation. The regulatory properties of pyruvate kinase M2 (PKM2) influence altered glucose metabolism in cancer.

Blood Lactate as a Prognostic Biomarker for Survival and Weight Loss in Amyotrophic Lateral Sclerosis: An Exploratory–Validation Study

open access: yesAnnals of Neurology, EarlyView.
Objective Lactate is increasingly recognized as an energy substrate, but its relevance to amyotrophic lateral sclerosis (ALS) remains unclear. We examined whether blood lactate is associated with survival and weight loss in ALS. Methods This retrospective study included an Australian exploratory cohort and a Japanese validation cohort with clinical ...

Research progress of PKM2-mediated regulatory pathways in periodontitis [PDF]

open access: yesKouqiang yixue
Enhanced glycolysis and other host metabolic reprogramming are recognized as core drivers of sustained inflammation. As a rate-limiting enzyme of glycolysis, the pyruvate kinase M2 (PKM2) exerts important effects on various chronic inflammatory diseases ...
