An integrated network approach identifies the isobutanol response network of Escherichia coli

open access: yesMolecular Systems Biology, 2009
Isobutanol has emerged as a potential biofuel due to recent metabolic engineering efforts. Here we used gene expression and transcription network connectivity data, genetic knockouts, and network component analysis (NCA) to map the initial isobutanol ...

Comparison of calculated and experimental isotope edited FTIR difference spectra for purple bacterial photosynthetic reaction centers with different quinones incorporated into the QA binding site.

open access: yesFrontiers in Plant Science, 2013
Previously we have shown that ONIOM type (QM/MM) calculations can be used to simulate isotope edited FTIR difference spectra for neutral ubiquinone in the QA binding site in Rhodobacter sphaeroides photosynthetic reaction centers.

Natural Products Inspired Scaffold Diversification Leads to Unnatural Molecular Warhead and Covalent Strategy to Modulating Protein Function through Electrophilic Bromine Transfer

open access: yesAdvanced Science, EarlyView.
We report a new thiolate‐reactive α,α‐gem‐dibromo lactam warhead that activates transcription factor Nrf2 and demonstrates anti‐inflammatory activities, which have implications in cancer, neurodegeneration, and cardiovascular diseases. RNA‐seq illuminated detailed transcriptional profiles, and chemical reactions with cysteine‐containing compounds ...

Self‐Assembling Hybrid Hydrogel Reprograms the Stromal Vascular Fraction to Treat Osteoarthritis

open access: yesAdvanced Science, EarlyView.
This study presents a bioinspired injectable hydrogel that enhances the therapeutic potential of stem cell‐rich stromal vascular fraction for treating osteoarthritis. By reprogramming cell behavior through epigenetic modulation, the hydrogel promotes cartilage regeneration and reduces joint damage in a rat model, offering a promising new approach for ...

Covalent Inhibition of SHMT2 by Gambogic Acid Induces Ferroptosis Through Mitochondrial Collapse in Triple‐Negative Breast Cancer

open access: yesAdvanced Science, EarlyView.
In this study, chemoproteomics combined with genetic and functional analyses was integrated to identify SHMT2 as a covalent and functional target of gambogic acid (GA) in triple‐negative breast cancer (TNBC). Further validation demonstrated that GA selectively modifies the Cys241 site of SHMT2, triggering mitochondrial dysfunction, activating the Nrf2 ...

Convenient Synthesis of 2-Thioimidazolone/Menadione Conjugates via a Two-Step Sequence Starting with Direct Amination of Menadione

open access: yesSynOpen, 2020
A convenient route to conjugates of 2-thiohydantoins and menadione with linkers of various chemical nature through a two-stage reaction sequence (direct amination of the menadione with a functional amine followed by modification of a functional group) is

Biomimetic Polymerization of Tellurocysteine: Breaking the Natural Amino Acid Radioprotection Limitation

open access: yesAdvanced Science, EarlyView.
Inspired by natural amino acid polymer‐melanin systems, this study strategically incorporated tellurocysteine to develop advanced radiation‐shielding materials. This approach transformed the primary interaction mechanism between melanin‐based materials and incident photons from Compton scattering to the photoelectric effect. Tellurocysteine‐polymerized

Bioadhesive Hydrogel With Polyphenol‐Armored Nanogene Rejuvenates Chondrocyte Senescence for Aged Osteoarthritis Therapy

open access: yesAdvanced Science, EarlyView.
An adhesive hydrogel depot with polyphenol‐armored nanogenes is developed for treating aged osteoarthritis. This system maintains miR‐140 bioactivity, prolongs its intra‐articular retention, and enhances transfection into chondrocytes, leading to the downregulation of senescence‐associated pathways.

Characterization and Trypanocidal Activity of a Novel Pyranaphthoquinone

open access: yesMolecules, 2017
Chagas disease is an endemic parasitic infection that occurs in 21 Latin American countries. New therapies for this disease are urgently needed, as the only two drugs available (nifurtimox and benznidazol) have high toxicity and variable efficacy in the ...
