Castration‐resistant prostate cancer cells are addicted to the high activity of cyclin‐dependent kinase 2

open access: yesMolecular Oncology, EarlyView.
We show that emergence of castration‐resistant prostate (CRPC) is associated with significant upregulation of cyclins that positively regulate cyclin‐dependent kinase 2 (CDK2) and concomitant downregulation of CDK4 cyclins. This renders CRPC cells dependent on the high activity of CDK2, and CDK2 inhibitors synergistically sensitize CRPC cells to both ...

Enhancing microsegregation during rapid directional solidification through ternary microalloying

open access: yesMaterials & Design
The nano-cellular dendritic microstructure formed during rapid directional solidification in powder bed fusion additive manufacturing creates unique properties such as simultaneous improvement in strength and ductility.

Mechanisms and therapeutic opportunities of the ribotoxic stress response in cancer

open access: yesMolecular Oncology, EarlyView.
Cancer cells' high translational demand creates opportunities to therapeutically target ribosome function. Ribosome stalling and collisions activate ZAKα and the ribotoxic stress response (RSR), which can trigger rapid, p53‐independent apoptosis in cancer.

Single-track investigation of rapid solidification conditions of Hastelloy-X Ni-based superalloy in laser powder-bed fusion and applicability of columnar equiaxed transition criteria

open access: yesVirtual and Physical Prototyping
To understand the applicability of the columnar-equiaxed transition (CET) criteria to the rapid cooling conditions unique to the powder-bed fusion (PBF) type additive manufacturing (AM) process, we investigated relationships between the microstructure ...

The VHL tumor suppressor at the crossroad of protein folding, aggregation, and cancer

open access: yesMolecular Oncology, EarlyView.
Mutations, environmental stress, and chaperone dysfunction can destabilize pVHL, promoting its conversion from the native folded state into amyloid‐like assemblies. This transition may contribute to protein storage, cell dormancy, survival, and drug resistance.

Microstructure formation of rapidly/sub-rapidly solidified Gd-Co-Ti immiscible alloys

open access: yesJournal of Materials Research and Technology
Solidification experiments were conducted for ternary Gd-Co-Ti immiscible alloys by copper mold casting. The impacts of the composition and cooling rate on the microstructure features and the properties were investigated.

Regulation of the lncRNA NEAT1 by p53‐ΔNp63 crosstalk modulates the DNA damage response and therapeutic efficacy in HNSCC

open access: yesMolecular Oncology, EarlyView.
In head and neck squamous cell carcinoma (HNSCC) p53 and p63 exert opposite roles on the transcription regulation of the lncRNA NEAT1. Under basal conditions, p53 levels are low and p63 represses NEAT1 expression. Upon genotoxic stress, p53 is rapidly induced, displacing p63 from the NEAT1 promoter leading to NEAT1 transcriptional activation and ...

Paclitaxel induces NM2‐dependent cellular contraction through GEF‐H1 dissociation from microtubules and RhoA/ROCK activation in cancer cells

open access: yesMolecular Oncology, EarlyView.
Taxanes are widely used chemotherapeutics whose effects on cellular mechanics remain poorly understood. We show that paclitaxel induces rapid cellular contraction by promoting GEF‐H1 dissociation from microtubules and non‐muscle myosin II activation through RhoA/ROCK.

Effects of solidification shrinkage on solute segregation and hot cracking sensitivity in liquid channel during columnar dendrite growth

open access: yesJournal of Materials Research and Technology
Hot cracking is the most prevalent defect in the rapid solidification process of alloys, which seriously restricts the mechanical properties of the build.
