Histone deacetylase 6 controls Notch3 trafficking and degradation in T-cell acute lymphoblastic leukemia cells [PDF]

open access: yes, 2018
Several studies have revealed that endosomal sorting controls the steady-state levels of Notch at the cell surface in normal cells and prevents its inappropriate activation in the absence of ligands.

A 3D Astrocyte Microenvironment Model Enables Rapid Ca2+‐Resolved Analysis and Therapeutic Modulation of Oxidative Neuroinflammation

open access: yesAdvanced Healthcare Materials, EarlyView.
This work presents ARC‐3D, a soft 3D model that recreates how brain support cells, called astrocytes, react to oxidative stress. The system visualizes rapid calcium changes and inflammatory signals, and shows how the drug KDS12025 can protect cells from damage. ARC‐3D offers a simple, reliable way to study early drivers of brain inflammation.

Notch1 and IL-7 Receptor Signalling in Early T-cell Development and Leukaemia

open access: yes, 2012
Notch receptors are master regulators of many aspects of development and tissue renewal in metazoans. Notch1 activation is essential for T-cell specification of bone marrow-derived multipotent progenitors that seed the thymus, and for proliferation and further progression of early thymocytes along the T-cell lineage.

Low‐Temperature Fabrication of Thymosin β4‐Loaded Soluble Microneedles to Promote Wound Healing by Specific Binding to Downregulated Immune Regulators Vsig4 and IL22rɑ2

open access: yesAdvanced Healthcare Materials, EarlyView.
Schematic diagram depicting the fabrication and application of thymosin β4 (Tβ4)‐loaded microneedle patches for wound treatment. The Tβ4 was loaded into chitosan (CS) and sucrose MNs under mild conditions (4°C, 65% relative humidity). The Tβ4 MN patch specifically binds to the downregulated immune regulators Vsig4 and IL22rα2, thereby accelerating ...

Upregulation of miR-150-5p alleviates LPS-induced inflammatory response and apoptosis of RAW264.7 macrophages by targeting Notch1

open access: yesOpen Life Sciences, 2020
Circulating miR-150-5p has been identified as a prognostic marker in patients with critical illness and sepsis. Herein, we aimed to further explore the role and underlying mechanism of miR-150-5p in sepsis.

Monoallelic Heb/Tcf12 Deletion Reduces the Requirement for NOTCH1 Hyperactivation in T-Cell Acute Lymphoblastic Leukemia

open access: yesFrontiers in Immunology, 2022
Early T-cell development is precisely controlled by E proteins, that indistinguishably include HEB/TCF12 and E2A/TCF3 transcription factors, together with NOTCH1 and pre-T cell receptor (TCR) signalling.

Estrogen receptor β–dependent Notch1 activation protects vascular endothelium against tumor necrosis factor α (TNFα)-induced apoptosis [PDF]

open access: yesJournal of Biological Chemistry, 2017
Unlike age-matched men, premenopausal women benefit from cardiovascular protection. Estrogens protect against apoptosis of endothelial cells (ECs), one of the hallmarks of endothelial dysfunction leading to cardiovascular disorders, but the underlying molecular mechanisms remain poorly understood.

Piezoelectric Stimulation of Neural Cells: Exploring the Synergistic Potential of Hybrid Scaffolds for Enhanced Regeneration

open access: yesAdvanced Materials Interfaces, EarlyView.
Hybrid piezoelectric scaffolds offer a promising route for Central Nervous System regeneration by combining structural and electrical cues to support neural stem cell growth. This review highlights their potential to overcome current challenges in neural tissue engineering by exploring porous hybrid materials, their biological interactions, and ...

Downregulating Notch counteracts KrasG12D-induced ERK activation and oxidative phosphorylation in myeloproliferative neoplasm. [PDF]

open access: yes, 2019
The Notch signaling pathway contributes to the pathogenesis of a wide spectrum of human cancers, including hematopoietic malignancies. Its functions are highly dependent on the specific cellular context. Gain-of-function NOTCH1 mutations are prevalent in

Strawberry Notch 1 Acts as a Transcriptional Regulator Driving Oncogenic Programs in Liver Carcinogenesis

open access: yesAdvanced Science, EarlyView.
This study reports that SBNO1 protein is upregulated in several cancer entities. SBNO1 protein interacts with the basal transcription factor TFIID via TAF4, enabling its recruitment to transcription start sites and the modulation of target gene expression.
