Evidence for a Low-Affinity Interleukin-3 Receptor

Growth Factors, 1990
Interleukin-3 (IL-3) regulates the proliferation of myeloid, erythroid, and lymphoid cells. Previous reports showed IL-3 binding restricted to a single high-affinity (Kd = 50-200 pM) site. Here, we demonstrate by equilibrium studies an additional binding site for IL-3 with lower apparent affinity (Kd = 5-20 nM). Furthermore, kinetic analysis shows that

Affinity isolation of the interleukin-3 surface receptor

Biochemical and Biophysical Research Communications, 1986
Interleukin-3 (IL-3) is a pluripotent hematopoietic growth factor which can stimulate both proliferation and differentiation of progenitor cells. Using the factor dependent NSF-60 and FDC-P1 cells, which possess specific, high affinity binding sites for 125I-IL-3 (i.e.

Functional Interaction of c-Kit Receptor and Interleukin-3 Receptor.

Blood, 2007
Abstract Hematopoiesis is regulated by a number of growth factors and cytokines, among which stem cell factor (SCF) plays a critical role for normal hematopoiesis. An important feature of SCF is its strong capacity to synergize with the cytokines including interleukin-3(IL-3), granulocyte-marcrophage colony-stimulation factor (GM-CSF ...

Receptors for Interleukin-3 (IL-3)

1995
Abstract The murine and human IL-3R o. subunit cDNAs (GenBank accession numbers X64534 and M74782) encode a protein of 396 and 378 amino acids including a putative signal peptide of 16 and 18 amino acids, an extracellular domain of 315 and 288 amino acids, a single transmembrane spanning segment of 24 and 20 amino acids, and a ...

Properties of the Murine Interleukin-3 Receptor

1990
Murine interleukin-3 (mIL-3) is a hemopoietic growth factor produced by activated T lymphocytes that stimulates the survival, proliferation and differentiation of pluripotent stem cells and a variety of committed myeloid and lymphoid progenitors (1). Because of its wide range of biological activities both in vitro and in vivo (2), it is thought to play

Cloning of an Interleukin-3 Receptor Gene: a Member of a Distinct Receptor Gene Family

Science, 1990
Interleukin-3 (IL-3) binds to its receptor with high and low affinities, induces tyrosine phosphorylation, an d promotes the proliferation and differentiation of hematopoietic cells. A binding component of the IL-3 receptor was cloned. Fibroblasts transfected with the complementary DNA bound IL-3 with a low affinity [dissociation constant (

Characterization of the interleukin 3 receptor.

Experimental hematology, 1990
A variety of homobifunctional crosslinking agents have been used to gain insight into the nature of the murine interleukin 3 (mIL-3) receptor. When [125I]mIL-3 was cross-linked to receptor sites on the surfaces of intact B6SUtA1 cells with disuccinimidyl suberate (DSS), sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) revealed the ...

Interleukin-3-associated ganglioside GD1a is induced independently of normal interleukin-3 receptor in murine myelogenous leukaemia NFS60 cells transfected with the interleukin-3 gene

Glycoconjugate Journal, 1996
The mechanism of interleukin-3 (IL-3) independent cell growth and of IL-3-associated ganglioside expression was analysed using the IL-3 dependent murine myelogenous leukaemia cell line NFS60-I7 and IL-3 gene-transfected sublines. The transfected cell lines showed autonomous cell growth, tumorigenicity, and IL-3 associated ganglioside GD1a expression in
