Variability of Botulinum Toxins: Challenges and Opportunities for the Future
Botulinum neurotoxins (BoNTs) are the most potent known toxins, and are therefore classified as extremely harmful biological weapons. However, BoNTs are therapeutic drugs that are widely used and have an increasing number of applications.

IMM‐H018, a dual IDO1/NE inhibitor, demonstrates potent therapeutic efficacy in sepsis by simultaneously targeting excessive inflammation and immune dysfunction. It prevents both primary and secondary sepsis through anti‐inflammatory, immune‐restoring, and renoprotective mechanisms, reducing organ damage, improving immune homeostasis, preserving kidney

The membrane‐active peptide Pep19‐2.5 reduces harmful inflammation by blocking activation of the NLRP3 inflammasome at trans‐Golgi network membranes. By targeting key membrane interactions, Pep19‐2.5 suppresses inflammatory IL‐1β production and alleviates allergic airway inflammation in mice, leading to reduced immune cell infiltration and improved ...

Structural Polymorphism of polyG Inclusions Revealed by In Situ Cryo‐Electron Tomography
Correlative cryo‐electron tomography in primary cortical neurons and NIID mouse brain tissue reveals that polyG inclusions are interconnected ribbon‐like assemblies rather than canonical amyloid fibrils. Multiple compartment‐specific ribbon states show distinct 26S proteasome accessibility, while cytoplasmic ribbons contact and deform ER‐like ...

Rapid Purification of Endotoxin-Free RTX Toxins
Cytolytic leukotoxins of the repeat in toxin (RTX) family are large proteins excreted by gram-negative bacterial pathogens through the type 1 secretion system (T1SS).

In bladder cancer, LRG1 binds to ANXA2 to trigger mitochondrial ROS‐dependent NETosis. This pathogenic cascade actively uncouples endothelial‐mural cell interactions, driving profound vascular destabilization. Consequently, targeting the LRG1‐ANXA2 axis attenuates the neutrophil burden and induces structural vascular normalization, offering a powerful ...

Subversion of Atypical Mucin Traps by a Spore‐Coat Effector Blocks Cellular Immunity in Drosophila
While the entomopathogenic fungus Metarhizium robertsii can evade insect humoral immunity, its subversion of cellular defenses is still elusive. We identify Eac1 as a spore‐coat protein essential for infecting drosophilids by targeting atypical mucins (Sgf1 and Sgf2), which can entrap spores in a gel matrix and thereby facilitate hemocyte encapsulation.

Many bacterial toxins exert their cytotoxic effects by enzymatically inactivating one or more cytosolic targets in host cells. To reach their intracellular targets, these toxins possess functional domains or subdomains that interact with and exploit ...
