Regioselectivity of dechlorination: Reductive dechlorination of polychlorobiphenyls by polymethylhydrosiloxane-alkali metal

Abstract Polychlorobiphenyls are completely dechlorinated to biphenyl by a polymethylhydrosiloxane-alkali metal system at ambient conditions. Dechlorination proceeded via the formation of a polychlorobiphenyl radical anion, which cleaved with a regioselectivity similar to that normally observed in nucleophilic substitution of polyhaloarenes.

Strategies and procedures for enriching, isolating, and cultivating reductively dechlorinating bacteria that use chloroorganic compounds as metabolic electron acceptors from environmental samples are described. Further, nucleic acid-based approaches used to detect and quantify dechlorinator (i.e., Dehalococcoides)-specific genes are presented.

Abstract: The sodium hydride based reducing agents were prepared the method of Caubere (Angew. Chem. Int. Ed. Engl., 1983, 22, 599-613). Reagents were prepared containing nickel, iron and zinc metals. All were effective in dechlorinating 1,3-dichlorobenzene, 4-methoxy-chlorobenzene, and arochlor 1254.

An anaerobic mixed bacterial culture was enriched for bacteria dechlorinating 1,2,3- and 1,2,4-trichlorobenzene (TCB) to dichlorobenzenes by exclusive use of non-fermentable substrates and the application of vancomycin. Growth and dechlorination occurred in a purely synthetic medium with formate or hydrogen, acetate, and TCB.
