Relative dose intensity of first-line triplet chemotherapy in metastatic colorectal cancer

open access: yesDigestive and Liver Disease
Relative dose intensity (RDI) is a measurement of chemotherapy (CT) dose defined as the actual dose received divided by the standard calculated dose during a set period. The study objective was to assess the impact of a RDI ≥ 80% on response and survival of patients treated in first line CT by FOLFOXIRI or FOLFIRINOX ± Bevacizumab (BV) for an ...

Influence of chemotherapy intensity on opportunistic fungal infection risk in non-small cell lung carcinoma: a retrospective study

open access: yesFrontiers in Medicine
BackgroundOpportunistic fungal infections represent a serious treatment-related complication in patients with advanced non-small cell lung carcinoma (NSCLC) undergoing chemotherapy, closely associated with treatment intensity-induced immunosuppression ...

Establishment of a humanized patient‐derived xenograft mouse model of high‐grade serous ovarian cancer for preclinical evaluation of combination immunotherapy

open access: yesMolecular Oncology, EarlyView.
We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...

CCDC80 suppresses high‐grade serous ovarian cancer migration via negative regulation of B7‐H3

open access: yesMolecular Oncology, EarlyView.
PAX8 is a lineage‐specific master regulator of transcription in high‐grade serous ovarian cancer (HGSC) progression. We show for the first time that PAX8 facilitates proliferation and metastasis by repressing the cell autonomous tumor suppressor CCDC80 and inducing B7‐H3 expression.

E2A selectively regulates TGF‐β–induced apoptosis in KRAS‐mutant non‐small cell lung cancer

open access: yesMolecular Oncology, EarlyView.
Ability to induce apoptosis by TGF‐β is frequently lost in advanced lung adenocarcinoma despite intact TGF‐β signaling. We identify E2A as a mutant KRAS–dependent mediator of resistance to TGF‐β–induced apoptosis. TGF‐β induces E2A via SMAD3 in mutant KRAS cells, and E2A silencing restores apoptosis and enhances radiation response in cell lines ...

CD47 promotes mitogen‐activated protein kinase and epithelial‐to‐mesenchymal transition molecular programs to drive prometastatic phenotypes in non‐small cell lung cancer

open access: yesMolecular Oncology, EarlyView.
Beyond its role in immune evasion, this study identified that CD47 drives tumor‐intrinsic signaling in non‐small cell lung cancer (NSCLC). Transcriptomic profiling and functional studies revealed that CD47 regulates cell adhesion, migration, and metastasis through an ERK–EMT signaling axis.

Should thrombopoietin receptor agonists be used for chemotherapy-induced thrombocytopenia?

open access: yesResearch and Practice in Thrombosis and Haemostasis
Chemotherapy-induced thrombocytopenia (CIT) is a common complication of cancer therapy for solid tumors that results in increased bleeding risk and chemotherapy dose reductions, treatment delays, and agent discontinuation.

KDM7A and KDM1A inhibition suppresses tumour promoting pathways in prostate cancer

open access: yesMolecular Oncology, EarlyView.
Treatment resistance is a major challenge for patients with advanced prostate cancer. This study examined an alternative approach to target the major prostate cancer‐promoting pathway by targeting epigenetic factors, whose levels are higher in tumours.

Optimizing Lenvatinib Therapy for Prognostic Improvement in Advanced Thyroid Cancer

open access: yesPharmaceuticals
Radioiodine-refractory differentiated thyroid cancer (RAIR-DTC) is associated with poor prognosis and limited systemic options. This narrative review focuses on lenvatinib (LEN), a multitarget tyrosine kinase inhibitor that significantly prolongs ...

The average relative dose intensity of R‐CHOP is an independent factor determining favorable overall survival in diffuse large B‐cell lymphoma patients

open access: yesCancer Medicine, 2019
The prognosis of diffuse large B‐cell lymphoma (DLBCL) patients depends on lymphoma‐ and patient‐related risk factors and is best estimated by the international prognostic index (IPI).
