Surface‐Capped Protein Nanoparticles for Nonviral Gene Delivery

open access: yesAdvanced Materials, EarlyView.
ABSTRACT Developing simple, safe, and efficient nonviral delivery systems remains a significant challenge in bioengineering. Nanoparticles offer promising gene delivery capabilities with reduced toxicity; however, long‐standing challenges related to effective plasmid encapsulation and delivery exist.

Super‐Enhancer‐Driven SOX4/SMAD3 Mediate Membrane Remodeling by Regulating Phospholipid Metabolism to Accelerate Leukemia Progression

open access: yesAdvanced Science, EarlyView.
Chronic myeloid leukemia blast phase (CML‐BP) poses a severe therapeutic challenge. This study reveals that the super‐enhancer‐driven transcription factors SOX4 and SMAD3 form a cooperative axis critical for disease progression. They co‐activate the oncogenic kinase AXL and promote phospholipid remodeling via LPCAT1 to facilitate its signaling ...

Inhibition of Calcium‐Dependent Lipid Droplets Relocation of ACSL4‐PKCβ‐ALOX15 Complex Alleviates Ferroptosis and Acute Pancreatitis

open access: yesAdvanced Science, EarlyView.
This study identifies L‐type calcium channel blockers as novel ferroptosis inhibitors. It reveals that PKCβ, activated in calcium dependent manner, phosphorylates and activates ACSL4 and ALOX15, relocating them to lipid droplets to promote lethal lipid peroxidation and ferroptosis.

Genetic landscape of Italian SRLV: insights from passive surveillance-based phylogenetic analysis

open access: yesFrontiers in Microbiology
Small ruminant lentiviruses (SRLVs) are retroviruses infecting small ruminants, characterized by a high genetic variability. SRLVs infection may result in a variety of clinical signs in goats and sheep and is not species-specific.

Human Endogenous Retroviruses in Neurological Diseases

open access: yesTrends in Molecular Medicine, 2018
The causes of multiple sclerosis and amyotrophic lateral sclerosis have long remained elusive. A new category of pathogenic components, normally dormant within human genomes, has been identified: human endogenous retroviruses (HERVs). These represent ∼8%

A Sister Lineage of Sampled Retroviruses Corroborates the Complex Evolution of Retroviruses

open access: yesMolecular biology and evolution, 2020
The origin and deep history of retroviruses remain mysterious and contentious, largely because the diversity of retroviruses is incompletely understood. Here, we report the discovery of lokiretroviruses, a novel major lineage of retroviruses, within the ...

GPCRs in CAR‐T Cell Immunotherapy: Expanding the Target Landscape and Enhancing Therapeutic Efficacy

open access: yesAdvanced Science, EarlyView.
Chimeric antigen receptor T cell therapy faces dual challenges of target scarcity and an immunosuppressive microenvironment in solid tumors. This review highlights how G protein‐coupled receptors can serve as both novel targets to expand the therapeutic scope and functional modules to enhance CAR‐T cell efficacy.

Viral Infections and Systemic Lupus Erythematosus: New Players in an Old Story

open access: yesViruses, 2021
A causal link between viral infections and autoimmunity has been studied for a long time and the role of some viruses in the induction or exacerbation of systemic lupus erythematosus (SLE) in genetically predisposed patients has been proved. The strength

BCR::ABL1‐Induced Enhancer Reprogramming Uncovers Hypersensitivity of Ph+B‐ALL Cells to Enhancer‐Targeting Drugs

open access: yesAdvanced Science, EarlyView.
Ng et al. show that the BCR::ABL1 kinase that drives the lymphoid leukemia Ph+B‐ALL modulates enhancer function by coopting signaling‐inducible transcription factors such as MYC, STAT5, and ETV5. BCR::ABL1 thereby promotes the transcriptional program driving and defining this leukemia and renders Ph+B‐ALL cells hypersensitive to enhancer‐inhibiting ...

Sensitive detection systems for infectious agents in xenotransplantation*

open access: yesXenotransplantation, EarlyView., 2020
Abstract Xenotransplantation of pig cells, tissues, or organs may be associated with transmission of porcine microorganisms, first of all of viruses, to the transplant recipient, potentially inducing a disease (zoonosis). I would like to define detection systems as the complex of sample generation, sample preparation, sample origin, time of sampling ...
