ERVmap analysis reveals genome-wide transcription of human endogenous retroviruses

open access: yesProceedings of the National Academy of Sciences of the United States of America, 2018
Significance Endogenous retrovirus (ERV) sequences make up a large fraction of our genome, yet little is understood about their function and biological relevance. Deep-sequencing data contain valuable information on a genome-wide scale. Yet, due to their

PD‐L1‐Binding Antigen Presenters: Redirecting Vaccine‐Induced Antibodies for Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
The PBAP‐gE complex anchors gE antigen to PD‐L1 on tumor cells. Vaccine‐induced anti‐gE antibodies simultaneously engage FcγRIIIa on NK cells and tumor‐bound PBAP‐gE, triggering NK cell activation and antibody‐dependent cellular cytotoxicity, thereby selectively eliminating PD‐L1–expressing tumor cells.

Retroviruses and Autoimmunity.

open access: yesInternal Medicine, 2001
The investigation of human retroviruses has shown dramatic progress since the discovery of human immunodeficiency virus (HIV). These studies have also contributed to the exploration of the role of retroviruses, including endogenous retroviruses, in the induction of autoimmune diseases such as systemic lupus erythematosus (SLE).

DADA Enhances CD8+ T Cell Stemness to Improve Anti‐Tumor Immunity and Immunotherapy Efficacy

open access: yesAdvanced Science, EarlyView.
Diisopropylamine dichloroacetate (DADA) enhances CD8+ T cell stemness by improving OXPHOS and mitochondrial fitness in a PDK1‐depenpendent manner. This metabolic shift strengthens CD8+ T cell anti‐tumor immunity, improves responses to PD‐1 blockade, and endows CAR‐T cells with prolonged persistence and resistance to terminal exhaustion, highlighting a ...

Inhibiting DNA Methylation Causes an Interferon Response in Cancer via dsRNA Including Endogenous Retroviruses.

open access: yesCell, 2015
We show that DNA methyltransferase inhibitors (DNMTis) upregulate immune signaling in cancer through the viral defense pathway. In ovarian cancer (OC), DNMTis trigger cytosolic sensing of double-stranded RNA (dsRNA) causing a type I interferon response ...

T Cell Exhaustion in Cancer Immunotherapy: Heterogeneity, Mechanisms, and Therapeutic Opportunities

open access: yesAdvanced Science, EarlyView.
T cell exhaustion limits immunotherapy efficacy. This article delineates its progression from stem‐like to terminally exhausted states, governed by persistent antigen, transcription factors, epigenetics, and metabolism. It maps the exhaustion landscape in the TME and proposes integrated reversal strategies, providing a translational roadmap to overcome

Retroviruses drive the rapid evolution of mammalian APOBEC3 genes

open access: yesProceedings of the National Academy of Sciences of the United States of America, 2019
Significance It is thought that evolution of antiviral genes has been shaped over the long term by antagonistic interactions with viruses, but in most cases this is challenging to investigate.

Targeting the Mapk13‐Tcf1‐Slc7a5 Axis via One‐Carbon Metabolic Regulation to Prevent Chronic Allograft Vasculopathy

open access: yesAdvanced Science, EarlyView.
This study emphasizes the role of the Mapk13‐Tcf1‐Slc7a5‐methionine metabolism axis in stem‐like CD4+ T cells. Moreover, it uncovers the mechanism through which limiting one‐carbon metabolism in CD4+ stem‐like T cells suppresses the tide of chronic allograft vasculopathy, offering potential targets to promote long‐term graft survival.

Human Endogenous Retroviruses and Type 1 Diabetes

open access: yesCurrent Diabetes Reports, 2019
The aim of this review is to discuss recent data pointing at an involvement of human endogenous retroviruses (HERVs) in type 1 diabetes (T1D) onset and progression.

Automatically Defining Protein Words for Diverse Functional Predictions Based on Attention Analysis of a Protein Language Model

open access: yesAdvanced Science, EarlyView.
Understanding protein sequence–function relationships remains challenging due to poorly defined motifs and limited residue‐level annotations. An annotation‐agnostic framework is introduced that segments protein sequences into “protein words” using attention patterns from protein language models.
