Condensin Promotes Position Effects within Tandem DNA Repeats via the RITS Complex

open access: yesCell Reports, 2016
Summary: Tandem repetitive DNA is highly abundant in eukaryotic genomes and contributes to transcription control and genome stability. However, how the individual sequences within tandem repeats behave remains largely unknown.

Pulmonary Delivery of siRNA Anti‐TNFα‐loaded Lipid Nanoparticles for Rapid Recovery in Murine Acute Lung Injury

open access: yesAdvanced Healthcare Materials, EarlyView.
In a murine model of acute lung injury, pulmonary administration of lipid nanoparticles carrying TNFα siRNA enables targeted delivery to immune cells. The study emphasises the importance of optimizing LNP distribution and delivery timing to improve therapeutic outcomes and develop emergency therapies for lung hyperinflammation.

Nanocarrier‐Based Targeting of Pattern Recognition Receptors as an Innovative Strategy for Enhancing Sepsis Therapy

open access: yesAdvanced Healthcare Materials, EarlyView.
This review highlights recent progress in nanocarriers targeting pattern recognition receptors (PRRs), including Toll‐like and NOD‐like receptors, for enhancing the treatment of bacterial sepsis and related complications. These nanomedicines deliver antibiotics and anti‐inflammatory agents while modulating immune responses.

Molecular Mechanisms of Extracellular Vesicle Biogenesis and Their Impact on the Design of Custom EVs

open access: yesAdvanced Healthcare Materials, EarlyView.
Extracellular vesicles (EVs) are pivotal mediators of intercellular communication and disease, yet the fundamental mechanisms controlling their biogenesis and cargo selection remain unclear. This limitation hinders their diagnostic utility and therapeutic development.

Gene up-regulation by DNA demethylation in 35S-gshI-transgenic poplars (Populus x canescens) [PDF]

open access: yes, 2008
Gene expression levels of transgene 35S-gshI (γ-glutamylcysteine synthetase) cloned from E. coli, and the endogenous gene gsh1 of poplar (Populus x canescens) were upregulated by the DNA demethylating agent DHAC (5,6-dihydro-5'-azacytidine ...

Suppressing t(4;11) Acute Leukemia by Lipopolymer Nanoparticle Delivery of siRNA Targeting KMT2A::AFF1 with Enhanced Extrahepatic Delivery

open access: yesAdvanced Healthcare Materials, EarlyView.
This study introduces a new lipopolymer nanoparticle (LPNP) system that efficiently delivers siRNA to leukemia cells. The LPNPs silence the leukemia fusion gene KMT2A::AFF1, induce apoptosis, and decrease leukemia burden in mice. These results demonstrate the potential of LPNPs as a targeted siRNA therapy for acute lymphoblastic leukemia.

Development of a Modular Ribonucleoprotein Complex as a General Strategy to Deliver RNAi Therapeutics

open access: yesAdvanced Healthcare Materials, EarlyView.
A modular and customizable RNAi therapeutics delivery platform composed of a pseudosymmetric human RNA binding protein‐siRNA complex (SmiRNP) is developed. By fusing a targeting module and an endosomal escape peptide onto the delivery platform, SmiRNP successfully delivered siRNA to silence the KRAS oncogene in a colorectal carcinoma cell line, leading
