Transcripts enriched in codons that trigger P‐site tRNA‐mediated mRNA decay possess stable mRNA

open access: yesFEBS Open Bio, EarlyView.
PTMD codons were first described by Mendel et al. as mediators of an mRNA decay pathway dependent on the human protein CNOT3, homologous to yeast Not5. Our findings confirm that PTMD codons destabilize transcripts; however, unlike in yeast, the human pathway specifically targets and slightly destabilizes primarily stable mRNAs.

Functional domains of the influenza A virus PB2 protein:identification of NP- and PB1-binding sites

open access: yes, 2004
Influenza virus genomic RNA segments are packaged into ribonucleoprotein (RNP) structures by the PB1, PB2, and PA subunits of an RNA polymerase and a single-strand RNA-binding nucleoprotein (NP).

A Split NanoLuc Reporter Quantitatively Measures Circular RNA IRES Translation

open access: yes, 2022
Internal ribosomal entry sites (IRESs) are RNA secondary structures that mediate translation independent from the m7G RNA cap. The dicistronic luciferase assay is the most frequently used method to measure IRES-mediated translation.

Cost analysis of residual viremia detected by two real-time PCR assays for response-guided (dual or triple) therapy of HCV genotype 1 infection

open access: yesGlobal & Regional Health Technology Assessment, 2016
Cost analysis of residual viremia detected by two real-time PCR assays for response-guided (dual or triple) therapy of HCV genotype 1 infectionIntroductionThe duration of interferon-based dual and triple therapies for HCV-G1 is determined by assessment ...

Microenvironmental Reprogramming by 3D Anisotropic Cardiac Extracellular Matrix Induces Nuclear Remodeling and Epigenetic Maturation of Chemically Induced Cardiomyocytes

open access: yesAdvanced Functional Materials, EarlyView.
A 3D anisotropic hydrogel derived from heart extracellular matrix guides cytoskeletal alignment and nuclear remodeling in reprogrammed cardiomyocyte‐like cells. This study reveals how matrix alignment modulates nuclear envelope dynamics and chromatin state, triggering transcriptional and functional maturation.

Role of RNA Interference (RNAi) in the Moss Physcomitrella patens [PDF]

open access: yes, 2013
RNA interference (RNAi) is a mechanism that regulates genes by either transcriptional (TGS) or posttranscriptional gene silencing (PTGS), required for genome maintenance and proper development of an organism.

Exploring an Alternative to mRNA Vaccine Cold Chain Storage: MRNA‐Lipid Nanoparticle Stability When Dried in a Polymer Matrix

open access: yesAdvanced Functional Materials, EarlyView.
The nanostructure, size, and function of mRNA‐loaded lipid nanoparticles are evaluated before drying, within polymer microneedles, and after rehydration. The results reveal the polymer and LNP loadings required to recover nanostructure and preserve the delivery performance in dry‐state formulations.

Activation of influenza virus RNA polymerase by the 5' and 3' terminal duplex of genomic RNA

open access: yes, 2003
The current model for influenza virus mRNA transcription involves the sequential interaction of the viral polymerase with the 5'- and 3'-ends of vRNA, with each RNA-protein interaction triggering a polymerase function necessary for cap-primed ...

Activation of the interferon induction cascade by influenza A viruses requires viral RNA synthesis and nuclear export

open access: yes, 2014
This work is supported by grants from the Wellcome Trust (grants 087751/A/08/Z) and MRC (G1001726/1).We have examined the requirements for virus transcription and replication and thus the roles of input and progeny genomes in the generation of interferon

A role for the Cajal-body-associated SUMO isopeptidase USPL1 in snRNA transcription mediated by RNA polymerase II [PDF]

open access: yes, 2014
Cajal bodies are nuclear structures that are involved in biogenesis of snRNPs and snoRNPs, maintenance of telomeres and processing of histone mRNA. Recently, the SUMO isopeptidase USPL1 was identified as a component of Cajal bodies that is essential for ...
