HROB Induces Lung Adenocarcinoma Progression via ZC3HC1‐CCNB1 Axis Regulation and Cell Cycle Dysregulation

We have uncovered a novel mechanism by which HROB modulates cell proliferation in lung adenocarcinoma. Our findings suggest that HROB recruits ZC3HC1 and mediates its de‐phosphorylation at Ser354, which in turn promotes K27‐linked ubiquitination of CCNB1, leading to its proteasomal degradation.

To identify genetic events that can lead to tumour death once either MLH1 or TP53 is mutated, a genome‐wide genetic screening was performed, uncovering a list of putative hits. Synthetic lethal interactions were validated either genetically or chemically by using small molecules that inhibit these genes either in vitro and in vivo.
