Dual PI3K/AKT and CDK4/6 inhibition reveals selective sensitivity in an SHH medulloblastoma stem cell model

open access: yesMolecular Oncology, EarlyView.
Targeted therapy was evaluated in SHH medulloblastoma using neuroepithelial stem cell (NES) and tumor‐derived NES‐like (tNES) models in 2D monolayers and 3D spheroids. PI3K, AKT, and CDK4/6 inhibitors had minimal effects in NES but markedly reduced viability and growth and induced apoptosis in tNES cells, revealing distinct therapeutic vulnerabilities.

Study on Composition Design and Performance Characteristics of Warm-Mixed Rubber–Asphalt Mixture for Cold-Region Stress Absorption Layers

open access: yesBuildings
Reflection cracks significantly compromise the service life of half-rigid asphalt pavements in cold regions. This study introduces SAKIII warm-mixed rubber–asphalt mixture (SAKIII WMRA Mix) as a stress absorption layer to address this issue.

KDM7A and KDM1A inhibition suppresses tumour promoting pathways in prostate cancer

open access: yesMolecular Oncology, EarlyView.
Treatment resistance is a major challenge for patients with advanced prostate cancer. This study examined an alternative approach to target the major prostate cancer‐promoting pathway by targeting epigenetic factors, whose levels are higher in tumours.

Developing sustainable road infrastructure performance indicators using a model-driven fuzzy spatial multi-criteria decision making method

open access: yes, 2020
Road infrastructure performance is closely associated with passengers and freight transportation systems and socio-economic development. The performance of road infrastructure is commonly measured by sensor-monitored indicators, and the ability of ...

Mechanical Properties of Sandstone Cement-Stabilized Macadam

open access: yesApplied Sciences, 2019
Application of sandstone in cement-stabilized macadam (CSM) is an effective way to utilize sandstone. To determine the feasibility of using sandstone as a CSM aggregate, a series of experimental investigations, such as unconfined compressive strength ...

Hijacking emergency granulopoiesis: Neutrophil ontogeny and reprogramming in cancer

open access: yesMolecular Oncology, EarlyView.
Neutrophils are highly plastic innate immune cells; their functions in cancer extend beyond the tumour microenvironment. This Review summarises current understanding of neutrophil maturation and heterogeneity and highlights tumour‐induced granulopoiesis as a systemic programme that expands immature, immunosuppressive neutrophils via tumour‐derived ...

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Metastasis on pause: How dormant tumor cells stay hidden within the tumor microenvironment and evade immune surveillance

open access: yesMolecular Oncology, EarlyView.
Dormant cancer cells can hide in distant organs for years, evading treatment and the immune system. This review highlights how signals from the surrounding tissue and immune environment keep these cells inactive or trigger their reawakening. Understanding these mechanisms may help develop therapies to eliminate or control dormant cells and prevent ...
