Chromatography of cephalosporins on DEAE-Sephadex

A sephadex extraction of urinary steroid conjugates

Abstract A simple method for the isolation of the C 19 and C 21 steroid conjugates from urine with Sephadex G-50 (coarse) is described. Following contraction of the urine volume by Sephadex, the Sephadex cake was washed with ethanol: methanoi (1:1 v/v) mixture and this was added to the contracted urine sample and evaporated to dryness.

Affinity of Mutarotase for Sephadex and Glycogen

It was found that hog kidney mutarotase has an affinity for Sephadex G-100, equilibrated and eluted with 5 mmol/l EDTA buffer (pH 7.4), resulting in retardation of its elution from the column. The affinity was reduced depending upon the presence of 0.15 mol/l NaCl and 0.2 mol/l glucose, separately or in combination.

The large scale syntheses (2-10 mmoles) of these starting blocks required chromatography through large columns of benzoylated DEAE-cellulose. This is a time-consuming and laborious step.
