Novel drugs approved by the EMA, the FDA and the MHRA in 2025: A year in review
Abstract In the 2025 novel drug mini‐review, one can take a full measure of the ingenuity that underlies current drug design and development, despite the year's smaller harvest (46 novel drugs) compared to 2024 (53) and 2023 (70). 54% of the novel drugs are first‐in‐class (FIC).

Effects of Sepiapterin Supplementation and NOS Inhibition on Glucocorticoid-Induced Hypertension
Glucocorticoid-induced hypertension is associated with imbalance between nitric oxide (NO) and superoxide. One of the pathways that causes this imbalance is endothelial NO synthase (eNOS) uncoupling.

Cloning And Ontogenic Expression Of Sepiapterin Reductase Mrna In Zebrafish Embryo : Possible Role In Pteridine Biosynthesis [QL767. K95 2008 f rb]. [PDF]

A fat-derived metabolite regulates a peptidergic feeding circuit in Drosophila. [PDF]
Here, we show that the enzymatic cofactor tetrahydrobiopterin (BH4) inhibits feeding in Drosophila. BH4 biosynthesis requires the sequential action of the conserved enzymes Punch, Purple, and Sepiapterin Reductase (Sptr).

ABSTRACT The correct diagnosis of tetrahydrobiopterin (BH4, sapropterin dihydrochloride)‐responsive phenylketonuria (PKU) and treatment with BH4 are important for prognosis and quality of life. We examined whether age affects biopterin bioavailability following oral BH4 administration in PKU and whether this influences BH4 responsiveness.

Brain Age in Adult Patients With Early‐Treated Phenylketonuria
ABSTRACT Structural brain alterations have been observed in individuals with phenylketonuria (PKU); however, the potential impact of PKU on brain aging remains unexplored. This study investigated brain age in adults with early‐treated classical PKU compared to healthy controls. Thirty early‐treated adults with classical PKU (age 19–48 years) and 59 age‐

L-sepiapterin restores SLE serum-induced markers of endothelial function in endothelial cells
Objective SLE serves as an independent risk factor` for endothelial dysfunction (ED) not explained by Framingham risk factors. We sought to understand the development of SLE-induced ED on a cellular level in order to develop strategies aimed at reversing

ABSTRACT Colour polytypism represents an example of phenotypic diversification shaped by genetic divergence and ecological pressures. Poison frogs of the genus Oophaga (Dendrobatidae) are highly polytypic in coloration, making them an ideal system for investigating the genetic and physiological basis of colour variation.

The purpose of this study was to characterize the protective effect of tetrahydrobiopterin (BH4), one of the cofactors of nitric oxide (NO) synthase, against NO-induced endothelial cell injury. The addition of S-nitroso-N-acetyl-D,L-penicillamine (SNAP),

Levodopa Use and Long‐Term Benefit in Different Dystonia Phenotypes
Background Dopaminergic changes have been reported in various forms of dystonia beyond dopa‐responsive dystonia (DRD). Besides DRD, levodopa (LDopa) is generally not considered effective in dystonia and may worsen it. This study assesses the impact of LDopa on different dystonia phenotypes. Objectives The patient database of Tampere University Hospital
