Background: One-carbon metabolism involves both mitochondrial and cytosolic forms of folate-dependent enzymes in mammalian cells, but few in vivo data exist to characterize the biochemical processes involved. Objective: We conducted a stable-isotopic ...

Combinations of small activating RNAs and small interfering RNAs were developed as personalized precision therapies that simultaneously activate tumor suppressors and silence oncogenes according to the molecular signatures of patient tumors. This combination strategy demonstrated superior anticancer efficacy, highlighting the promise of precision ...

Effect of oxygen tension on the amino acid utilisation of human embryonic stem cells
Background/aims: human embryonic stem cells (hESCs) are a potential source of cells for treatment of many degenerative diseases, but in culture have a propensity to spontaneously differentiate, possibly due to suboptimal conditions. Culture at low oxygen

Repurposing a Small Molecule Plant Hormone as a Tunable ON‐Switch for CAR‐T Cell Immunotherapy
By engineering a receptor system integrating the plant auxin receptor AFB1 with its co‐receptor IAA7, we enable ligand‐dependent interactions triggered by the plant hormone auxins. This design allows rapid, reversible, and dose‐dependent T cell activation, resulting in potent cytotoxicity against B‐cell lymphoma in vitro and in vivo.

D-serine is an endogenous N-methyl-D-aspartate (NMDA) receptor coagonist. It is synthesized from L-serine by serine racemase (SRR), but many aspects of its metabolism remain unclear, especially in the forebrain, which lacks active D-amino acid oxidase ...

Objective: Tumor-associated macrophages (TAMs) exhibit heterogeneous properties including anti-tumorigenic and pro-tumorigenic phenotypes. The rate-limiting enzyme in de novo serine biosynthesis, 3-phosphoglycerate dehydrogenase (PHGDH), has a well ...

Fabrication and use of D-serine biosensors for characterising D-serine signalling in rat brain [PDF]
D-serine is a co-agonist at NMDA receptors in the brain but the study of this amino acid is restricted by current techniques. I have designed highly sensitive D-serine biosensors that permit accurate real-time recordings of D-serine in the brain in a ...

In this study, MPDA NPs were engineered to encapsulate CTT. To further increase renal accumulation of CTT, L‐serine was conjugated to the surface of the MPDA nanoparticles (CTT/MPDA@L‐Ser NPs). CTT/MPDA@L‐Ser NPs inhibit DPEP1 activity, thereby restoring ferroptosis defense mechanisms through upregulation of GPX4 and SLC7A11 expression, ultimately ...
