The complexity of serotype replacement of pneumococci [PDF]

open access: yesHuman Vaccines & Immunotherapeutics, 2019
Pneumococci respond extremely rapidly to the vaccine pressure created by the conjugate vaccines (PCVs). Vaccine serotypes are disappearing, meanwhile new, often previously very rare types are emerging, and it is difficult to establish what makes certain serotypes more successful in replacement.

Transcriptomic functional characterization of recombinant adeno‐associated virus producing cell line adapted to suspension‐growth

open access: yesBiotechnology Progress, EarlyView.
Abstract Recombinant adeno‐associated virus (rAAV) is a widely used delivery vehicle in gene therapy. A scalable production technology is essential for its wide clinical applications. We have taken a synthetic biology approach to generate HEK293‐based cell lines which harbor integrated genetic elements encoding essential AAV and adenoviral helper ...

Accelerating AAV purification process development using high‐throughput resin tip module

open access: yesBiotechnology Progress, EarlyView.
Abstract Recombinant adeno‐associated viruses (AAVs) with precise genome editing and cell‐virus interaction have become a promising delivery tool for gene therapy. A robust AAV purification process is crucial for ensuring therapeutic efficacy. The challenges of AAV purification process development encompass limited material availability during early ...

Harvesting AAV by tangential flow filtration using reverse asymmetric membranes

open access: yesBiotechnology Progress, EarlyView.
Abstract Efficient bioreactor clarification for harvesting virus particles is often challenging. Tangential flow filtration is attractive as it can be easily adapted for batch and perfusion operations. Here the feasibility of using reverse asymmetric hollow fiber membranes, where the more open support structure faces the feed stream, has been ...

Reactivity of Carbohydrate Phosphodiesters, Potential Targets of Antibacterial Agents

open access: yesChemistry &Biodiversity, EarlyView.
A simple carbohydrate phosphodiester model was synthesized to compare the reactivity of phosphodiester bonds within carbohydrates and RNA. ABSTRACT While monosaccharide units in bacterial carbohydrates may be linked with phosphodiester bonds, human carbohydrates exclusively contain glycosidic linkages.
