A Spotlight on Yolk‐sac Tumors: Molecular Pathology, Current Diagnostics, and Novel Therapeutics

open access: yesAndrology, EarlyView.
ABSTRACT Background Yolk‐sac tumors are an aggressive subtype of testicular cancer that significantly contribute to disease progression and therapy resistance, especially in adults. While testicular cancer generally has high cure rates with cisplatin‐based treatment, adult yolk‐sac tumors often appear as components of mixed tumors with poor response to

Inhibition of miR-143-3p Restores Blood–Testis Barrier Function and Ameliorates Sertoli Cell Senescence

open access: yesCells
Due to the increasing trend of delayed childbirth, the age-related decline in male reproductive function has become a widely recognized issue. Sertoli cells (SCs) play a vital role in creating the necessary microenvironment for spermatogenesis in the ...

Intrauterine Growth and Male Reproductive Health: A Follow‐up Study From the Danish National Birth Cohort

open access: yesAndrology, EarlyView.
ABSTRACT Background Reduced intrauterine growth might affect male reproductive health later in life, however, existing research remain inconclusive. Objectives To investigate the association between placental weight and birthweight Z‐scores and semen characteristics, testes volume, and reproductive hormone levels in young men.

Variants in ZZS Complex‐Associated Genes TEX11 and M1AP Are Responsible for Male Infertility and Nonobstructive Azoospermia

open access: yesAndrology, EarlyView.
ABSTRACT Background Nonobstructive azoospermia (NOA) is the most severe form of male infertility, with genetic factors contributing to approximately 30% of cases. However, only a small fraction of all NOA cases can be explained by the current genetic findings.

Can Hormonal Therapy Improve the Outcomes of mTESE in Patients With Non‐Obstructive Azoospermia?

open access: yesAndrology, EarlyView.
ABSTRACT Background Non‐obstructive azoospermia (NOA) represents the most severe form of male infertility. Hypogonadism is common in NOA patients, and normal testosterone (T) levels are considered essential for spermatogenesis. Fertility‐preserving hormonal therapy (FpHT) has been proposed to optimize hormonal milieu and improve sperm retrieval rates ...

Machine Learning‐Based Prediction of Sperm Retrieval Outcomes in Patients With Klinefelter Syndrome: A Multicenter Study With External Validation

open access: yesAndrology, EarlyView.
ABSTRACT Background The Klinefelter syndrome is a common genetic cause of male infertility, and testicular sperm extraction (TESE) enables sperm retrieval in a subset of affected patients. However, predicting TESE success remains challenging due to the heterogeneous clinical and endocrinological presentation of the Klinefelter syndrome.

Menopausal hormone therapy after ovarian cancer: A 10‐year survival analysis in premenopausal women

open access: yesActa Obstetricia et Gynecologica Scandinavica, EarlyView.
Among premenopausal women surgically treated for ovarian cancer, no difference in 10‐year overall survival was observed between postoperative MHT users and non‐users. Young ovarian cancer survivors should be recommended MHT after surgically induced menopause.

MTAP Deficiency as a Metabolic Vulnerability in Cancer: Implications for Synthetic Lethal Therapy

open access: yesCancer Science, EarlyView.
MTAP deletion creates a therapeutically actionable metabolic vulnerability through MTA accumulation and PRMT5 dependency. This review summarizes the biochemical basis of MTAP‐directed synthetic lethality, emerging PRMT5/MAT2A inhibitors, clinicogenomic features of MTAP‐deleted tumors, and future strategies for precision oncology.

Pubertal Dynamics of Sertoli and Leydig Cell Dysfunction in Klinefelter Syndrome

open access: yesClinical Endocrinology, EarlyView.
ABSTRACT Context Klinefelter syndrome (KS), defined by a 47, XXY karyotype, is commonly associated with progressive testicular failure. The precise timing of Sertoli and Leydig cell dysfunction during puberty remains unclear. Objective To determine the onset and progression of testicular insufficiency during puberty in KS, and to assess whether ...

A novel classification of testicular sex cord‐stromal tumours from the Testicular Sex Cord‐Stromal Tumour (TESST) group: a collaboration of the Genitourinary Pathology Society (GUPS) and the International Society of Urological Pathology (ISUP)

open access: yesHistopathology, EarlyView.
The TESST classification of testicular sex cord‐stromal tumours was agreed upon by consensus based on the best available evidence. The picture shows the members of the TESST group and delegates of GUPS and ISUP present in‐person at the first meeting of the group held at Johns Hopkins Hospital (Baltimore, 2024). Aims Testicular sex cord‐stromal tumours (
