The Structural Basis of Pathogenic Subgenomic Flavivirus RNA (sfRNA) Production [PDF]

open access: yesScience, 2014
Resisting the Chop Dengue, West Nile, and Yellow Fever viruses are all flaviviruses that have single-stranded RNA genomes and form specific, short flaviviral RNAs (sfRNAs) during infection that cause viral pathogenicity. These sfRNAs are produced by the incomplete degradation of viral RNA by the host-cell exonuclease Xrn1.

Flavivirus sfRNA suppresses antiviral RNA interference in cultured cells and mosquitoes and directly interacts with the RNAi machinery

open access: yesVirology, 2015
Productive arbovirus infections require mechanisms to suppress or circumvent the cellular RNA interference (RNAi) pathway, a major antiviral response in mosquitoes. In this study, we demonstrate that two flaviviruses, Dengue virus and Kunjin virus, significantly repress siRNA-mediated RNAi in infected human cells as well as during infection of the ...

Promoter-like activity on the minus strand supports sfRNA biogenesis in Japanese encephalitis virus

Virology
Arthropod-borne flaviviruses produce subgenomic RNAs (sfRNAs) from the highly conserved 3'-untranslated region (UTR). While most flaviviruses generate sfRNAs by resisting degradation from the host exoribonuclease XRN1, Japanese encephalitis virus (JEV) maintains sfRNA production even when XRN1 is depleted, suggesting an alternative mechanism.

Multiple orthoflaviviruses secrete sfRNA in mosquito saliva to promote transmission by inhibiting MDA5-mediated early interferon response

open access: yesNature Communications
Abstract Numerous orthoflaviviruses transmitted through the bites of different mosquito species infect more than 500 million people annually. Skin infection at the bite site represents a critical and conserved step in transmission and a deeper understanding of this process will promote the design of broad-spectrum ...

Diferentes orthoflavivirus producen sfRNA que promueve la transmisión viral al inhibir la respuesta interferón

open access: yesSalud Uninorte
