Oxadiazole-carbonylaminothioureas as SIRT1 and SIRT2 Inhibitors

Journal of Medicinal Chemistry, 2008
A new inhibitor for human sirtuin type proteins 1 and 2 (SIRT1 and SIRT2) was discovered through virtual database screening in search of new scaffolds. A series of compounds was synthesized based on the hit compound (3-[[3-(4-tert-butylphenyl)1,2,4-oxadiazole-5-carbonyl]amino]-1-[3-(trifluoromethyl)phenyl]thiourea).

Tenovin-1 inhibited dengue virus replication through SIRT2

European Journal of Pharmacology, 2021
Dengue fever is a common arbovirus disease, which has been spread to the entire tropical world. At present, effective drugs for the treatment of dengue fever have not yet appeared, and the dengue vaccines studied in various countries have also experienced severe adverse reactions. Thus it is urgent to find new chemicals against dengue virus.

SIRT2 regulates microtubule stabilization in diabetic cardiomyopathy

European Journal of Pharmacology, 2015
Stable microtubules (MTs) is involved the mechanism of diabetic cardiomyopathy (DCM), which is induced by acetylation of α-tubulin. The present study investigated whether SIRT2, a deacetylase, regulates MT stability through α-tubulin deacetylation in DCM and whether the receptor of advanced glycation end products (AGEs) signaling pathway is involved in

HBx-elevated SIRT2 promotes HBV replication and hepatocarcinogenesis

Biochemical and Biophysical Research Communications, 2018
Sirtuin 2 (SIRT2) is a class III histone deacetylase that has been implicated to promote HCC development. However, the functional role of SIRT2 in HBV is still unclear. In this study, we found that HBV could upregulate SIRT2 expression. Additionally, HBx could activate SIRT2 promoter to upregulate the mRNA and protein level of SIRT2.

New SIRT2 inhibitors: Histidine-based bleomycin spin-off

Bioorganic & Medicinal Chemistry, 2019
Bleomycin is considered to exert its antitumor activity via DNA cleavage mediated by activated oxygen generated from the iron complex in its chelator moiety. Spin-offs from this moiety, HPH-1Trt and HPH-2Trt, with anti-cancer activities were recently synthesized.

SIRT2 Mediated Microtubule Acetylation in Osteogenic Differentiation.

The Chinese journal of dental research
To assess the role of microtubule acetylation in the transportation of amorphous calcium phosphate (ACP)-containing vesicles that mediate the osteogenic differentiation process of rat bone mesenchymal stem cells (BMSCs).Rat BMSCs were cultured and transfected with sirtuin 2 (SIRT2) overexpression plasmids for an in vitro model.

Structure of the histone deacetylase SIRT2.

Nature structural biology, 2001
Sir2 is an NAD-dependent histone deacetylase that mediates transcriptional silencing at mating-type loci, telomeres and ribosomal gene clusters, and has a critical role in the determination of life span in yeast and Caenorhabditis elegans. The 1.7 A crystal structure of the 323 amino acid catalytic core of human SIRT2, a homolog of yeast Sir2, reveals ...

Role of microglial SIRT2 on neuroinflammation

Sirtuin 2 (SIRT2) has been associated with aging, age-related pathologies and neuroinflammation. In the present study, we have analyzed in mice the implication of microglial sirtuin 2 in two different models of neuroinflammation: an acute pro-inflammatory stimulus (intraperitoneal injection of LPS) and neurodegeneration (mouse model of Alzheimer ...
