Early‐onset colorectal cancer (EOCRC) presents a complex molecular landscape that requires integrated multi‐omics approaches. This article reviews current applications of systems genetics in EOCRC research and provides a step‐by‐step protocol for network‐based analyses featuring PIK3CA to demonstrate practical application for researchers of all levels.

Hepatocyte TrkB is identified as a critical gatekeeper against MASH‐related fibrosis. Mechanistically, TrkB inhibits the TGFβ/SMAD3/FOS axis to suppress CCL2 secretion, thereby blocking pathogenic macrophage recruitment and ameliorating liver fibrosis.

Alpha‐crystallin B chain (CRYAB) has been reported to stabilize mothers against decapentaplegic homolog 4 (SMAD4) in transforming growth factor‐β (TGF‐β) signaling, enabling target gene transcription. We show nuclear CRYAB–SMAD4 interaction for the first time in human skeletal muscle fibers and its regulation by exercise.
