Feasibility and Tolerability of Performing Portable MRI for Neurological Disorders in an Outpatient Neurology Clinic: A Prospective Cohort

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Background Accessing brain magnetic resonance imaging (MRI) can be challenging, especially for underserved patients, which may lead to disparities in neurological diagnosis. Method This mixed‐methods study enrolled adults with one of four neurological disorders: mild cognitive impairment or dementia of the Alzheimer type, multiple sclerosis ...

Smoothened determines β-arrestin–mediated removal of the G protein–coupled receptor Gpr161 from the primary cilium

open access: yesJournal of Cell Biology, 2016
Pal et al. describe a two-step process determining removal of the cilia-localized GPCR, Gpr161, upon sonic hedgehog signaling. First, β-arrestins are recruited by the signaling-competent receptor in a smoothened-dependent manner.

Smoothened is a poor prognosis factor and a potential therapeutic target in glioma

open access: yes, 2017
Malignant gliomas are associated with a high mortality rate. Thus, there is an urgent need for the development of novel targeted therapeutics. Aberrant Hedgehog signaling has been directly linked to glioma. GDC-0449 is a novel small molecule inhibitor of

Secreted tyrosine kinase Vlk negatively regulates Hedgehog signaling by inducing lysosomal degradation of Smoothened

open access: yes, 2020
Vlk is a secreted tyrosine kinase that plays crucial roles during vertebrate embryonic development including skeletal formation. Genetic studies suggest that Vlk can modulate the Hedgehog signaling pathway during skeletal development.

Temporal and spatial requirements of Smoothened in ventral midbrain neuronal development [PDF]

open access: yes, 2013
Background Several studies have indicated that Sonic hedgehog (Shh) regulates the expansion of dopaminergic (DA) progenitors and the subsequent generation of mature DA neurons.

Fluid Biomarkers of Disease Burden and Cognitive Dysfunction in Progressive Supranuclear Palsy

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective Identifying objective biomarkers for progressive supranuclear palsy (PSP) is crucial to improving diagnosis and establishing clinical trial and treatment endpoints. This study evaluated fluid biomarkers in PSP versus controls and their associations with regional 18F‐PI‐2620 tau‐PET, clinical, and cognitive outcomes.

Pancreatic Islet-Autonomous Insulin and Smoothened-Mediated Signaling Modulate Identity Changes of Glucagon+ α-Cells

open access: yesNature Cell Biology, 2018
The mechanisms that restrict regeneration and maintain cell identity following injury are poorly characterized in higher vertebrates. Following β-cell loss, 1–2% of the glucagon-producing α-cells spontaneously engage in insulin production in mice.

Structure–Function Decoupling of the Sensorimotor and Default Mode Networks in Black Americans With MS

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Background and Objectives Multiple sclerosis (MS) exhibits racially disparate rates of disease progression. Black people with MS (B‐PwMS) experience a more severe disease course than non‐Hispanic White people with MS (NHW‐PwMS). Here we investigated structural and functional connectivity as well as structure–function decoupling in the ...

mTORC1 hampers Hedgehog signaling in Tsc2 deficient cells

open access: yesLife Science Alliance
This is the first demonstration that mTORC1 inhibits Hedgehog signaling. mTORC1 prevents cilia accumulation of Smo and activation of GlI2. Also, evidence is provided for a 2-step activation process of GLI2.
