Echogenic liposome as a carrier of siRNA for sonoporation: An alternative microbubble for sonoporation

2015 IEEE International Ultrasonics Symposium (IUS), 2015
Ultrasound contrast agents (UCAs), which are groups of engineered microbubbles, have been recently studied for drug delivery applications since cavitation of bubbles can increase the temporary permeability of nearby cells. However, the internal volume of UCAs is generally filled with gas, hence loading drug molecules into UCAs is limited.

Sonoporation of erythrocytes by lithotripter shockwaves in vitro

Ultrasonics, 1998
Sonoporation of red blood cells was examined in relation to cavitation-induced hemolysis. FITC-dextran at 580,000 MW was added to suspensions of canine erythrocytes and the mixture was exposed to lithotripter shockwaves. Exposure at 5% or 50% hematocrit in PBS or 50% in plasma yielded not only hemolysis but also FITC-dextran uptake in surviving cells ...

Theoretical study of microbubble dynamics in sonoporation

Ultrasonics, 2015
Sonoporation is a promising technology for promoting the transfer of drug or gene into cells using ultrasound-mediated microbubbles that transiently break up the cell membrane. In this article, a model is established to analyze the dynamics of ultrasound-mediated microbubble near the cell membrane, which may be especially useful for understanding the ...

Introduction of Genes via Sonoporation and Electroporation

2014
Gene therapy delivery using viral vectors has demonstrated efficient transfection but has safety issues. The need for safer yet effective delivery systems has led to the active development of non-viral techniques. In this chapter, we will discuss two evolving techniques - sonoporation with microbubble contrast agents and electroporation with focus on ...

Sonoporation of Cells for Drug and Gene delivery

The 26th Annual International Conference of the IEEE Engineering in Medicine and Biology Society, 2005
Recent studies demonstrate that ultrasound can be used to deliver compounds into viable cells for potential targeted drug delivery and non-viral gene transfection, revealing new, advantageous possibilities. The delivery is facilitated through sonoporation, the formation of temporary pores in the cell membrane induced by ultrasound. Our study focuses on

Sonoporation, drug delivery, and gene therapy

Proceedings of the Institution of Mechanical Engineers, Part H: Journal of Engineering in Medicine, 2009
Ultrasound is a very effective modality for drug delivery and gene therapy because energy that is non-invasively transmitted through the skin can be focused deeply into the human body in a specific location and employed to release drugs at that site. Ultrasound cavitation, enhanced by injected microbubbles, perturbs cell membrane structures to cause ...

Sonoporation of endothelial cells in vivo

2011 IEEE International Ultrasonics Symposium, 2011
Ultrasound contrast agents as drug delivery systems are an emerging field. Recently, it was shown that targeted microbubbles (tMB) are able to sonoporate endothelial cells in vitro. This study investigates if tMB can also induce sonoporation in vivo, thereby making it possible to combine molecular imaging and drug delivery.

Sonoporation in Developmental Biology

2009
Recent, molecular biology techniques have accomplished a large contribution to developmental biology. Especially, gene transduction techniques are indispensable to study the roles of regulatory genes underlying embryogenesis. Replication-competent retroviruses, transfection with lipofection and an in ovo electroporation have been established as gene ...

Noninvasive vaccination: From the perspective of sonoporation

Vaccine, 2008
Previous noninvasive vaccine submission approaches including respiratory tract-based methods are limited to specific pathogens. Recent developments on ultrasound biology suggest that ultrasound-mediated molecule submission (sonoporation) could be an effective noninvasive approach for gene therapy as well as drug submission.
