Castration‐resistant prostate cancer cells are addicted to the high activity of cyclin‐dependent kinase 2

open access: yesMolecular Oncology, EarlyView.
We show that emergence of castration‐resistant prostate (CRPC) is associated with significant upregulation of cyclins that positively regulate cyclin‐dependent kinase 2 (CDK2) and concomitant downregulation of CDK4 cyclins. This renders CRPC cells dependent on the high activity of CDK2, and CDK2 inhibitors synergistically sensitize CRPC cells to both ...

Investigation of Effecting Parameters in a Turning Operation

open access: yesAl-Khawarizmi Engineering Journal, 2019
In this study multi objective optimization is utilized to optimize a turning operation to reveal the appropriate level of process features. The goal of this work is to evaluate the optimal combination of cutting parameters like feed, spindle speed ...

An Improved Underdamped Asymmetric Bistable Stochastic Resonance Method and its Application for Spindle Bearing Fault Diagnosis

open access: yesIEEE Access, 2020
High-precision spindle bearing is one of the most critical and vulnerable parts in a motorized spindle. Its unexpected failure may lead to production loss.

Dynamic modeling and chatter analysis of a spindle-workpiece-tailstock system for the turning of flexible parts [PDF]

open access: yesThe International Journal of Advanced Manufacturing Technology, 2019
Chatter happens frequently in turning of flexible parts, jeopardizing the dynamic stability of the operation. The chatter stability prediction is dependent on the accurate estimation of the structural parameters of the cutting system. This paper aims to quantify the influence of the support conditions on dynamics of the flexible parts in straight ...

METHOD OF SELECTING THE OPERATION PARAMETERS OF ELECTRIC SPINDLE WITH AEROSTATIC SUPPORTS FOR SEPARATION OF SEMICONDUCTOR PLATES TO CRYSTALS. PART 1. DEVELOPMENT OF A MATHEMATICAL MODEL OF AXIAL OSCILLATIONS OF THE ELECTRIC SPINDLE [PDF]

open access: yesMechanics of Machines, Mechanisms and Materials, 2020
The article presents the results of studies of the mechanism of the occurrence of axial vibrations in the mechanical system “electrospindle shaft with aerostatic bearings – cutting diamond disk – cuttable semiconductor wafer”. A mathematical model of the resulting axial vibrations of the electrospindle shaft is proposed, taking into account the natural

Paclitaxel induces NM2‐dependent cellular contraction through GEF‐H1 dissociation from microtubules and RhoA/ROCK activation in cancer cells

open access: yesMolecular Oncology, EarlyView.
Taxanes are widely used chemotherapeutics whose effects on cellular mechanics remain poorly understood. We show that paclitaxel induces rapid cellular contraction by promoting GEF‐H1 dissociation from microtubules and non‐muscle myosin II activation through RhoA/ROCK.

Kinesin 6 Regulation in Drosophila Female Meiosis by the Non-conserved N- and C- Terminal Domains

open access: yesG3: Genes, Genomes, Genetics, 2018
Bipolar spindle assembly occurs in the absence of centrosomes in the oocytes of most organisms. In the absence of centrosomes in Drosophila oocytes, we have proposed that the kinesin 6 Subito, a MKLP-2 homolog, is required for establishing spindle ...

Dual pathway spindle assembly increases both the speed and the fidelity of mitosis [PDF]

open access: yes, 2011
Roughly half of all animal somatic cell spindles assemble by the classical prophase pathway, in which the centrosomes separate ahead of nuclear envelope breakdown (NEBD).

Engineering IL‐4 resistant proinflammatory human myeloid cells for cancer immunotherapy

open access: yesMolecular Oncology, EarlyView.
We developed a scalable workflow to generate proinflammatory human myeloid cells. CRISPR/Cas9‐edited CD34+ hematopoietic stem and progenitor cells were expanded and differentiated with M‐CSF. Deletion of STAT6 or STAT6/NFKB1 enhanced macrophage proinflammatory gene expression and cytokine secretion in the presence of IL‐4 while maintaining antibody ...

Tumour‐proximal deep serum proteomics identifies biomarkers associated with ovarian cancer

open access: yesMolecular Oncology, EarlyView.
New biomarkers are needed for early ovarian cancer detection. Ovarian (tumour‐proximal) and peripheral vein serum from healthy women and patients with benign or malignant tumours underwent nanoparticle‐enriched deep proteomics. Ovarian serum yielded more protein identifications and revealed tumour‐reflective signatures.
