Single‐Cell and Spatial Transcriptomic Profiling of Penile Squamous Cell Carcinoma Reveals Dynamics of Tumor Differentiation and Immune Microenvironment

open access: yesAdvanced Science, EarlyView.
This study uses single‐nucleus RNA sequencing and spatial transcriptomics to investigate penile squamous cell carcinoma (PSCC). It reveals that PSCC tumor cells mimic normal penile epithelium differentiation, independent of HPV status. The Tum_1 subtype shows basal stem‐like characteristics and promotes invasiveness. HPV‐positive basal stem‐like tumors

Organization of the Proteostasis Network of Membraneless Organelles

open access: yesAdvanced Science, EarlyView.
A comprehensive mapping of the proteostasis network (PN) across membraneless organelles (MLOs) reveals how central PN components coordinate MLO regulation. The analysis uncovers principles of specificity and promiscuity within the PN. The findings suggest that HSP70 chaperones act as generic MLO regulators, while client specific HSP70 co‐chaperones may

PRMT5 Inhibitor Synergizes with Chemotherapy to Induce Resembling Mismatch Repair Deficiency and Enhance Anti‐TIGIT Therapy in Microsatellite‐Stable Colorectal Cancer

open access: yesAdvanced Science, EarlyView.
Microsatellite‐stable (MSS) colorectal cancer (CRC) resists immunotherapy. PRMT5 inhibition enhances CPT‐11 sensitivity, inducing a PMS2‐deficient‐like state and cytosolic dsDNA release, which activates the cGAS‐STING pathway to promote anti‐tumor immunity. However, this combination therapy upregulates TIGIT expression on CD8+ T cells, and the addition

A Dual Approach with Organoid and CRISPR Screening Reveals ERCC6 as a Determinant of Cisplatin Resistance in Osteosarcoma

open access: yesAdvanced Science, EarlyView.
This study utilizes patient‐derived osteosarcoma organoids and CRISPR screening to identify ERCC6 and HNRNPM as key mediators of cisplatin resistance via the PI3K/AKT pathway and BAX alternative splicing. These findings provide novel insights and potential therapeutic targets for overcoming chemoresistance in osteosarcoma.
