SQSTM1 and Paget’s Disease of Bone

Calcified Tissue International, 2004
Mutations in the Sequestosome 1 gene ( SQSTM1; also known as p62) have recently been identified as the cause of 5q35-linked Paget's disease of bone (PDB). All of the mutations identified to date affect the ubiquitin-associated (UBA) domain of SQSTM1, a region of the protein that binds noncovalently to ubiquitin.

p62/SQSTM1 analysis in frontotemporal lobar degeneration

open access: yesNeurobiology of Aging, 2015
Mutations in the gene p62/SQSTM1 have been reported as a relatively rare cause of frontotemporal lobar degeneration (FTLD). To establish whether this was the case for cases of FTLD from the United Kingdom, we sequenced the sequenced the entire open reading frame of this gene in a large cohort of patients.

Defining the landscape of TIA1 and SQSTM1 digenic myopathy

open access: yesNeuromuscular Disorders
TIA1/SQSTM1 myopathy is one of the few digenic myopathies. We describe four new French adult male patients carrying the TIA1 p.Asn357Ser and SQSTM1 p.Pro392Leu variant and review the literature to include 20 additional cases to define the spectrum of the disease.

The role of SQSTM1 (p62) in mitochondrial function and clearance in human cortical neurons [PDF]

open access: yesStem Cell Reports, 2021
Sequestosome-1 (SQSTM1/p62) is involved in cellular processes such as autophagy and metabolic reprogramming. Mutations resulting in the loss of function of SQSTM1 lead to neurodegenerative diseases including frontotemporal dementia.

RIPosomes are targets of IRGM-SQSTM1-dependent autophagy

Autophagy, 2023
The NOD1-NOD2-RIPK2-NFKB/NF-κB pro-inflammatory axis plays a significant role in regulating the immune response to bacterial infection. However, an excess of NFKB-dependent cytokine response can be detrimental and, thus, should be kept under control to maintain the innate immune balance.

SQSTM1 is a therapeutic target for infection and sterile inflammation

Cytokine, 2023
Inflammation represents a fundamental immune response triggered by various detrimental stimuli, such as infections, tissue damage, toxins, and foreign substances. Protein degradation plays a crucial role in regulating the inflammatory process at multiple levels.

Regulation of selective autophagy: the p62/SQSTM1 paradigm

Essays in Biochemistry, 2017
In selective autophagy, cytoplasmic components are selected and tagged before being sequestered into an autophagosome by means of selective autophagy receptors such as p62/SQSTM1. In this review, we discuss how selective autophagy is regulated. An important level of regulation is the selection of proteins or organelles for degradation.

Use of p62/SQSTM1 antibodies for neuropathological diagnosis

Neuropathology and Applied Neurobiology, 2007
The demonstration of proteinaceous inclusions in the brain is the key step in the pathological diagnosis of degenerative dementias. The diversity of these diseases has necessitated the use of a panel of (immuno)stains to visualize all suspect pathologies, elevating diagnostic costs.
